Opportunities for developing innovative and intelligent drug delivery technologies by targeting the endocannabinoid system are becoming more apparent. This review provides an overview of strategies to develop targeted using (ECS). Recent advances in showcase enhanced pharmaceutical therapy specificity while minimizing undesirable side effects overcoming formulation challenges associated with cannabinoids. identifies that may permit access full pharmacotherapeutic potential ECS. The design optimized nanocarriers target specific tissues can be improved understanding nature signaling pathways, distribution mammalian body, receptor structure, enzymatic degradation A closer look at ligand-receptor complexes, tone, tissue distribution, G-protein activity leads a better ECS toolkit therapeutics. signal transduction pathways examine modulation downstream effector proteins, desensitization, cascades, biased signaling. An in-depth overall view is achieved through homology modeling where mutagenesis ligand binding site allow sequence analysis formation libraries molecular docking dynamic simulations. Internalization routes exploring receptor-mediated endocytosis lipid rafts also considered explicit Furthermore, highlights nanotechnology surface modification aspects as possible future approach targeting.

Some of the most prevalent neurodegenerative disorders, including Alzheimer’s and Parkinson’s disease, are proteinopathies characterized by accumulation specific protein aggregates in brain. Such misfolded can trigger modulation innate adaptive immune systems subsequently lead to chronic neuroinflammation that drives onset progression diseases. Since there is still no effective disease-modifying treatment, new therapeutic targets for have been sought. The endocannabinoid system, particular cannabinoid CB2 receptors, extensively studied, due their important role neuroinflammation, especially microglial cells. Several studies shown promising effects receptor activation on reducing aggregation-based pathology as well attenuating inflammation several dementia-related symptoms. In this review, we discuss available data receptors potential benefits limitations agonists these therapy proteinopathies.

Behaviour is the response of living things to their environment and external stimulation, one parameters be observed when assessing animal welfare. Any alteration from conditions found in nature can lead occurrence some specific behaviours, called stereotypies which are characterised as repetitive, consistent patterns behaviour usually defined having no apparent ultimate or proximal functions. It has been reported that once stabled subjected stressful activities, horses have more susceptibility developing behavioural disturbances; therefore, disorders a strong indicator poor Cannabis spp.-derived molecules studied under different medical conditions; therapeutic potentials phytocannabinoids related effects delta-9-tetrahydrocannabinol, cannabidiol (CBD), other compounds. Cannabidiol many activities within central nervous system, such anxiolytic, antidepressant, antipsychotic, anticonvulsant, anti-inflammatory activities. Some studies recently shown potential successful use veterinary medicine. This clinical case report described 22-year-old mare suffering chronic crib-biting wind-sucking, outcome four weeks-therapy with CBD. first equine disorders.

The implications of maternal overnutrition on offspring metabolic and neuroimmune development are well-known. Increasing evidence now suggests that obesity poor dietary habits during pregnancy lactation can increase the risk central peripheral dysregulation in offspring, but mechanisms not sufficiently established. Furthermore, despite many studies addressing preventive measures targeted at mother, very few propose practical approaches to treat damages when they already installed. Here we investigated potential cannabidiol (CBD) treatment attenuate effects induced by a cafeteria diet hypothalamic inflammation profile Wistar rats.We have observed range imbalances sex-dependant manner, with higher deposition visceral white adipose tissue, increased plasma fasting glucose lipopolysaccharides (LPS) levels both sexes, serum cholesterol triglycerides only occurred females, while insulin homeostatic model assessment index (HOMA-IR) was male offspring. We also found an overexpression pro-inflammatory cytokines tumor necrosis factor-alpha (TNFα), interleukin (IL) 6, 1β hypothalamus, trademark neuroinflammation. Interestingly, expression GFAP, marker for astrogliosis, reduced obese mothers, indicating adaptive mechanism utero Treatment 50 mg/kg CBD oil oral gavage able reduce tissue revert resistance males, LPS TNFα IL6 hypothalamus sexes.Together, these results indicate intricate interplay between counterparts pathogenicity therapeutic CBD. In this context, impairment internal circuitry caused neuroinflammation runs tandem disruptions important processes, which be attenuated ends.
