Ancistrobrevinium A (1) is the first N-methylated and non-hydrogenated, thus cationic naphthylisoquinoline alkaloid. It was discovered in root bark extract of phytochemically productive West African liana Ancistrocladus abbreviatus (Ancistrocladaceae). Its constitution elucidated by HR-ESI-MS 1D 2D NMR. Due to steric hindrance proximity linkage between naphthalene isoquinoline parts, biaryl axis rotationally hindered. constitutes a stable element chirality – only one new alkaloid since, different from most other alkaloids, it has no stereogenic centers. The axial configuration 1 assigned electronic circular dichroism (ECD) investigations, which gave positive couplet, indicating 'positive chirality', here corresponding P-configuration. showed weak cytotoxic activity against A549 lung cancer cells (IC50 = 50.6 μM).

Naphthylisoquinoline (NIQ) alkaloids are rising as a promising class of secondary metabolites with pharmaceutical potential. NF-κB has already been recognized significant modulator cancer proliferation and drug resistance. We have previously reported the mechanisms behind cytotoxic effect dioncophylline A, an NIQ monomer, in leukemia cells. In current study, we investigated jozimine A2, dimer, on cells comparison to second, structurally unsymmetric michellamine B. To this end, molecular docking was applied predict binding affinity dimers towards NF-κB, which then validated through microscale thermophoresis. Next, cytotoxicity assays were performed CCRF-CEM multidrug-resistant CEM/ADR5000 following treatment.
