Abstract Organohalide respiration (OHR), catalysed by reductive dehalogenases (RDases), plays an important role in halogen cycling. Natural organohalides and putative RDase-encoding genes have been reported Aarhus Bay sediments, however, OHR has not experimentally verified. The in vitro biochemical characterization revealed that iron/2-oxoglutarate (Fe/2OG)-dependent aliphatic halogenase WelO5* Hapalosiphon welwitschii IC-52-3 has an enhanced substrate specificity towards 12-epi-hapalindole C (1) comparison to WelO5 H. UTEX B1830. This allowed us define the origin of varied chlorinated versus dechlorinated alkaloid structural diversity between two welwitindolinone producers. Furthermore, this study, along with recent AmbO5 protein, collectively confirmed presence a signature sequence motif C-terminus newly discovered enzyme family confers promiscuity and specificity. Halogenated natural products (HNPs) are considered to be emerging contaminants whose environmental distribution and fate only incompletely known. Therefore, several persistent bioaccumulative HNP groups, together with man-made polychlorinated biphenyls (PCBs) polybrominated diphenyl ethers (PBDEs), were quantified in the blubber of nine sperm whales (Physeter macrocephalus) stranded on coast Mediterranean Sea Italy. The naturally occurring hexahydroxanthene derivatives (PBHDs; sum TetraBHD TriBHD) most prominent substance class up 77,000 ng/g blubber. mean PBHD content (35,800 blubber) even exceeded one PCBs (28,400 blubber), although region is known highly contaminated contaminants. Based values, Q1 ∼ PBDEs > MeO-BDEs 2,2′-diMeO-BB 80 other HNPs followed decreasing amounts.
