In Vivo Efficacy of Olorofim against Systemic Scedosporiosis and Lomentosporiosis

Clinically
relevant

resistant
against
currently
available
systemic
antifungal
agents
in
vitro,
infection
due
to
these

studied
vivo
efficacy

new
fungicidal
agent,
olorofim
(formerly
F901318),
scedosporiosis
lomentosporiosis
neutropenic
animals.
Cyclophosphamide-immunosuppressed
CD-1
mice
infected
by
Scedosporium
apiospermum,
Pseudallescheria
boydii
(Scedosporium
boydii),
were
treated
intraperitoneal
administration
(15
mg/kg
body
weight
every
8
h
for
9
days).
The
treatment
was
assessed
survival
rate
at
10
days
postinfection,
levels
serum
(1-3)-β-d-glucan
(BG),
histopathology,
fungal
burdens
kidneys
3
postinfection.
Olorofim
therapy
significantly
improved
compared
that
untreated
controls;
80%,
100%,
100%
survived
boydii,
prolificans,
respectively,
while
less
than
20%
control
(phosphate-buffered
saline
[PBS]
treated)
In
olorofim-treated
with
any
three
species,
BG
suppressed
DNA
detected
target
organs
lower
controls.
Furthermore,
histopathology
revealed
no
or
only
few
lesions
hyphal
elements
mice,
numerous
hyphae
present
mice.
These
results
indicate
be
promising
therapeutic
agent
scedosporiosis/lomentosporiosis,

emerging
infections

Further Support of Conspecificity of Oak and Mango Powdery Mildew and First Report of Erysiphe quercicola and Erysiphe alphitoides on Mango in Mainland Europe

Mango

multigene
sequencing
(ITS

coding
genes)
to
identify
the
causal
agent.
Erysiphe
quercicola
was
detected
97%
out
of
140
samples,
collected
six
different
orchards
Malaga
region.
Among
these,
a
small
proportion
also
yielded
E.
alphitoides
(8%
all
samples)
found
alone
3%
samples.
A
phylogenetic
approach
completed
cross
inoculations
between
oak
mango,
which
led
typical
symptoms,
supporting
conspecificity
mango
mildews.
To
our
knowledge,
this
is
first
report
causing
on
trees
mainland
Spain,
thus
Europe,
based
unequivocal
biological
evidence.

broad

range
