Background: We sought to replicate our 2015 findings linking chemical intolerance in parents with the risk of their children developing autism and/or ADHD. Drawing upon 2021 discovery a strong association between and mast cells, we propose an explanation for this link. Methods: In population-based survey U.S. adults, used internationally validated Quick Environmental Exposure Sensitivity Inventory (QEESI) assess symptom severity intolerance. Parents were asked how many biological had been diagnosed Results: scores top versus bottom tenth percentile 5.7 times reporting child 2.1 Conclusions: High among autism, coupled cell activation as plausible biomechanism intolerance, suggest that (1) QEESI can identify individuals at increased risk, (2) environmental counseling may reduce personal exposures (3) global rise ADHD be due fossil-fuel-derived biogenic toxicants epigenetically “turning on” or off” critical genes transmitted transgenerationally. Background Drug-induced thrombocytopenia (DITP) often occurs in patients during clinical treatment. However, clinicians usually fail to distinguish which drugs can be plausible culprits accurately. We aimed develop a large comprehensive drug benchmark database with DITP toxicity using the recommended method by FDA.

Assessing chemical safety is essential to evaluate the potential risks of exposure human health and environment. Traditional methods relying on animal testing are being replaced by 3R (reduction, refinement, replacement) principle-based alternatives, mainly depending in vitro test Adverse Outcome Pathway framework. However, these approaches often focus properties compound, missing broader chemical-biological interaction perspective. Currently, lack comprehensive molecular characterization system results limited real-world representation contextualization toxicological effect under study. Leveraging omics data strengthens understanding responses different biological systems, emphasizing holistic interactions when developing methods. Here, we discuss relevance meticulous two assessment relevant scenarios how omics-based, data-driven can improve future generation alternative

Abstract The development of a rapid and accurate model for determining the genotoxicity carcinogenicity chemicals is crucial effective cancer risk assessment, it also contributes to prevention. This study aims develop one-day, single-dose identifying genotoxic hepatocarcinogens (GHCs) in rats. Microarray gene expression data from livers rats administered single dose 58 compounds, including 5 GHCs, was obtained Open TG-GATEs database used identification marker genes construction predictive classifier identify GHCs We identified 10 markers commonly responsive all them construct support vector machine-based classifier. effectively distinguishes other demonstrating 100% sensitivity over 96% specificity. To further assess model’s effectiveness reliability, we conducted multi-institutional one-day oral administration studies on These examined 64 23 with via quantitative PCR (qPCR) 24 hours after administration. Our results demonstrate that qPCR analysis an alternative microarray analysis. GHC showed high accuracy achieving 91% (21/23) specificity 93% (38/41) across multiple validation three institutions. In conclusion, present proves be reliable highly sensitive tool anticipated valuable screening potential GHCs.

Abstract Omics techniques have been increasingly recognized as promising tools for Next Generation Risk Assessment. Targeted metabolomics offer the advantage of providing readily interpretable mechanistic information about perturbed biological pathways. In this study, a high-throughput LC–MS/MS-based broad targeted system was applied to study nitrofurantoin metabolic dynamics over time and concentration provide mechanistic-anchored approach point departure (PoD) derivation. Upon exposure at five concentrations (7.5 µM, 15 20 30 µM 120 µM) four points (3, 6, 24 48 h), intracellular metabolome HepG2 cells evaluated. total, 256 uniquely identified metabolites were measured, annotated, allocated in 13 different metabolite classes. Principal component analysis (PCA) univariate statistical showed clear metabolome-based effects. Mechanistic evidenced differential activation cellular pathways indicative early adaptive hepatotoxic response. At low concentrations, effects seen mainly energy lipid metabolism, mid range, antioxidant response by increased levels glutathione (GSH) from de novo GSH synthesis pathway. highest depletion GSH, together with alternations reflective mitochondrial impairments, Finally, metabolomics-based PoD derived multivariate PCA using whole set measured metabolites. This allows entire dataset derive that can be mechanistically anchored established key events. Our results show suitability high throughput investigate mechanisms hepatoxicity departures linked existing adverse outcome contribute development new ones.
