RNA 5-methylcytosine (m5C) modification has emerged as an important regulatory mechanism in the progression of human cancers, including hepatobiliary tumors. The m5C "reader" Aly/REF export factor (ALYREF) was recently found to be identified a prognostic biomarker liver cancer. However, its exact role intrahepatic cholangiocarcinoma (ICC) is unclear. In this study, ALYREF upregulated ICC tissues and cells. gain- loss-of-function experiments indicated that promoted cell proliferation invasion suppressed apoptosis. Moreover, we isocitrate dehydrogenase 1 (IDH1), metastatic marker cancer, also tissues, displayed relatively strong positive correlation with level ALYREF, positively regulated by ALYREF. As "reader", interacted m5C-IDH1 mRNA increased stability. knockdown partially eliminated promotion IDH1 on invasion. NRF2-driven glutathione synthesis cells, which reversed silencing. Finally, xenograft tumor mouse model, or treatment ivosidenib (an inhibitor) significantly growth vivo. conclusion, promotes increasing levels m5C-dependent manner.
