The neurobiological mechanisms that regulate the development and maintenance of alcohol use disorder (AUD) are complex involve a wide variety within between systems neuroadaptations. While classic reward, preoccupation, withdrawal neurocircuits have been heavily studied in terms AUD, viable treatment targets from this established literature not proven clinically effective as yet. Therefore, examination additional neurocircuitries classically context AUD may provide novel therapeutic targets. Recent studies demonstrate various neuropeptides important modulators seeking, intake behaviors. This includes neurocircuitry dorsal vagal (DVC), which is involved control autonomic nervous system, natural rewards like food, acts relay interoceptive sensory information via interactions numerous gut-brain peptides neurotransmitter with DVC projections to central peripheral neuron subtypes produce transmitters project target brain regions critical for reward such mesolimbic dopamine system well other limbic areas negative reinforcing aversive properties extended amygdala. suggests play role modulation aspects AUD. review summarizes current on neurotransmitters (e.g., norepinephrine, glucagon-like peptide 1, neurotensin, cholecystokinin, thyrotropin-releasing hormone), their potential relevance alcohol-related behaviors humans rodent models research. Abstract Alcohol use disorder (AUD) affects transcriptomic, epigenetic and proteomic expression in several organs, including the brain. There has not been a comprehensive analysis of altered protein abundance focusing on multiple brain regions that undergo neuroadaptations occurring AUD. We performed quantitative using liquid chromatography-tandem mass spectrometry (LC-MS/MS) human postmortem tissue from play key roles development maintenance AUD, amygdala (AMG), hippocampus (HIPP), hypothalamus (HYP), nucleus accumbens (NAc), prefrontal cortex (PFC) ventral tegmental area (VTA). Brain tissues were adult males with AUD ( n = 11) matched controls 16). Across two groups, there >6000 proteins quantified differential compared to each six regions. The region greatest number differentially expressed was AMG, followed by HYP. Pathways associated between groups (fold change > 1.5 LIMMA p < 0.01) analyzed Ingenuity Pathway Analysis (IPA). In adrenergic, opioid, oxytocin, GABA receptor cytokine pathways among most enriched. HYP, dopaminergic signaling Proteins highlight potential therapeutic targets such as CSNK1D (PF-670462), B opioid receptors may lead identification other targets. These results improve our understanding molecular alterations across are Proteomic data this study is publicly available at www.lmdomics.org/AUDBrainProteomeAtlas/ .
