Abstract In the ‘incubation of cocaine craving’ model relapse, rats exhibit progressive intensification (incubation) cue-induced craving over several weeks forced abstinence from self-administration. The expression incubated depends on plasticity excitatory synaptic transmission in nucleus accumbens core (NAcC) medium spiny neurons (MSN). Previously, we found that maintenance this and incubation ongoing protein translation, regulation translation is altered after craving. Here used male female express Cre recombinase either dopamine D1 receptor- or adenosine 2a (A2a) receptor-expressing MSN to a GFP-tagged ribosomal cell-type specific manner, enabling us use Translating Ribosome Affinity Purification (TRAP) isolate actively translating mRNAs both subtypes for analysis by RNA-seq. We compared self-administered saline cocaine. Saline were assessed day (AD) 1, while AD1 AD40-50. For D1-MSN A2a-MSN, there few differentially translated genes between groups. contrast, pronounced differences translatome observed AD40-50, was far more robust D1-MSN. Notably, all comparisons revealed sex mRNAs. Sequencing results validated qRT-PCR interest. This study, first combine TRAP-seq, transgenic rats, self-administration paradigm, identifies linked A2a-MSN NAcC.

Relapse to drug use is often preceded by craving. Clinical observations in the 1980s led clinical investigators postulate that cue-induced cocaine craving may increase during abstinence. Over 2 decades ago, identified an analogous phenomenon rats of time-dependent increases drug-seeking behavior homecage abstinence and termed it incubation In 2011, we reviewed first decade studies on brain mechanisms this review, provide update incubation-related from published since 2011. We review using standard method after homecage-forced cocaine, methamphetamine, opioid drugs, nicotine.
