ABSTRACT The rise of the Isthmus Panama separated populations many marine organisms, which then diverged into new geminate sister species currently living in Eastern Pacific Ocean and Caribbean Sea. However, we know very little about how such evolutionary divergences host have shaped compositions their microbiomes. Here, compared microbiomes whole-body shell-surface samples gastropods genera Cerithium Cerithideopsis to those congeneric outgroups. Our results suggest that effects ~3 million years separation isolation on microbiome composition varied among between sample types within same hosts. In samples, pairs tended be similar, likely due filtering, although strength this relationship two groups across similarity metrics. Shell-surface show contrasting patterns, with co-divergence taxa a small number microbial clades evident but not . Conceptual biogeographic frameworks have proposed that the relative contribution of environmental and geographical factors on microbial distribution depends several characteristics habitat (e.g. heterogeneity, species diversity, proportion specialist/generalist taxa), all them defining degree specificity, but few experimental demonstrations exist. Here, we aimed to determine effect specificity bacterial patterns assembly processes in benthic coastal ecosystems Southern Ocean (Patagonia, Falkland/Malvinas, Kerguelen, South Georgia, King George Islands), using 16S rRNA gene metabarcoding. The gradient resulted from a 'natural design' provided by Abatus sea urchin model, sediment (least specific habitat) intestinal tissue (most habitat). phylogenetic composition communities showed clear differentiation site, driven similar geographic distances. However, strength this pattern decreased with increasing specificity: stronger divergence compared gut tissue. stochastic deterministic contributing varied according scale level.
