Hypoxia,
a
state
of
reduced
oxygen
availability,
exerts
complex
and
often
paradoxical
effects
on
the
heart.
While
chronic
hypoxia
is
detrimental
leads
to
adverse
cardiac
remodeling
dysfunction,
short-term
or
intermittent
can
contribute
towards
protective
adaptations
that
enhances
heart's
ability
protect
itself
from
ischemic
injury.
This
adaptation,
also
known
as
hypoxic
preconditioning,
drives
activation
several
essential
signaling
pathways,
including
hypoxia-inducible
factor
(HIF)
signaling,
reactive
species
(ROS)
nitric
oxide
(NO)
regulation,
ATP-sensitive
potassium
channel
(KATP)
mediated
leading
metabolic
reprogramming,
angiogenesis
with
vascular
remodeling,
enhanced
calcium
handling,
thereby
increasing
resistance
heart
disease.
We
discuss
strategies
such
conditioning
pharmacologically
activated
HIF
along
targeted
approaches
enhance
glycolysis
KATP
regulation
optimize
sarcoplasmic
reticulum
ATPase
2a
(SERCA2a)
handling
in
cardiomyocytes.
In
this
review,
we
explore
mechanisms

Ammonia

Glycolytic
reprogramming
