Abstract This study systematically explores the oncogenic role of long non-coding RNA (lncRNA) LINC00115 in endometrial cancer (EC) and reveals its unique mechanism promoting proliferation, invasion, metastasis via JAK/STAT signaling pathway. is significantly upregulated EC tissues closely associated with advanced TNM staging lymph node metastasis. Background Disulfidptosis and ferroptosis are two different programmed cell death pathways, their potential therapeutic targets have important clinical prospects. Although there is an association between the two, role of genes associated with these forms in development endometrial cancer remains unclear. Methods In this study, RNA sequencing (RNA-seq) data were obtained from public databases, comprehensive analysis methods, including difference analysis, univariate Cox regression, Least Absolute Shrinkage Selection Operator (LASSO) used to construct a disulfidptosis/ferroptosis-related (DFRGs) prognostic signature. To further explore new feature, pathway functional analyses performed, differences gene mutation frequency level immune infiltration high- low-risk groups studied. Finally, we validated expression profile samples. Results We identified five optimal DFRGs that differentially expressed prognosis uterine corpus carcinoma (UCEC). These include CDKN2A, FZD7, LCN2, ACTN4, MYH10. Based on DFRGs, constructed robust model significantly lower overall survival high-risk group than group, tumor burden invasion risk groups. The key genes, ACTN4 was verified by immunohistochemistry RT-qPCR. Conclusion This study established characteristics assessment disulfidptosis provides insights guide patient management personalized treatment.

Endometrial cancer (UCEC) is the most prevalent gynecological malignancy in high-income countries, and its incidence rising globally. Although early-stage UCEC can be treated with surgery, advanced cases have a poor prognosis, highlighting need for effective molecular biomarkers to improve diagnosis prognosis. In this study, we analyzed mRNA miRNA sequencing data from tissues adjacent non-cancerous TCGA database. Differential expression analysis was conducted using DESeq2 package, identifying differentially expressed lncRNAs, miRNAs, mRNAs (DElncRNAs, DEmiRNAs, DEmRNAs). Key molecules were screened LASSO regression, ceRNA network constructed by predicting lncRNA-miRNA miRNA-mRNA interaction, which visualized Cytoscape. Functional enrichment elucidated roles mechanisms of network. The prognostic potential identified RNAs assessed through survival Cox regression analyses, while methylation immune infiltration analyses explored regulatory interactions. We lncRNA-miRNA-mRNA UCEC, centered on CDKN2B-AS1-hsa-miR-497-5p-IGF2BP3 axis. Survival confirmed significance network, univariate demonstrating strong association between aberrant overall prognosis UCEC. However, multivariate suggested that other clinical factors may modulate relationship. Methylation revealed low levels IGF2BP3, possibly contributing overexpression. Furthermore, studies highlighted significant correlations CDKN2B-AS1, multiple cell types, suggesting axis regulates tumor microenvironment. These findings suggest key element therapeutic target.

Abstract Background Long non-coding RNAs (lncRNAs) are crucial regulators of ovarian cancer(OC), playing a significant role in malignant transformation and closely linked to poor prognosis. Therefore, it is investigate the impact lncRNAs on biological behavior cancer understand their underlying molecular mechanisms. Methods The binding sites target genes were predicted through bioinformatics analysis, gene expression levels measured using qRT-PCR. cells was assessed cell function assays. Gene targeting relationships verified dual-luciferase reporter gene(DLRG) assay. Protein analyzed Western blotting. Results Hyaluronan-mediated motility receptor antisense RNA1(HMMR-AS1) upregulated cells. Cell experiments demonstrated that HMMR-AS1 promotes behaviors DLRG targets miR-627-3p, which turn pleiotrophin(PTN). Furthermore, we discovered functions as competing endogenous RNA (ceRNA) for regulating PTN thereby promoting phenotypes Conclusions In summary, our study indicates lncRNA highly expressed plays carcinogenic role. Targeting may offer novel therapeutic strategy treating cancer.
