Abstract Non-suicidal self-injury (NSSI) is a transdiagnostic psychiatric symptom with high prevalence and relevance in child adolescent psychiatry. Therefore, it of great interest to identify biological phenotype associated NSSI. The aim the present study was cross-sectionally investigate patterns markers underlying NSSI psychopathology sample female patients healthy controls. Comprehensive clinical data, saliva blood samples, heart rate variability pain sensitivity, were collected n = 149 40 participants. Using machine-based learning, we tested extent which oxytocin, dehydroepiandrosterone (DHEA), beta-endorphin, free triiodothyronine (fT3), leukocytes, sensitivity able classify participants regarding their outcomes NSSI, depression borderline personality disorder symptomatology. We evaluated predictive performance several models (linear logistic regression, elastic net random forests, gradient boosted trees) using repeated cross-validation. With as an outcome variable, both regression machine learning showed moderate (Area under Receiver Operating Characteristic Curve between 0.67 0.69). Predictors highest power low oxytocin (OR 0.55; p 0.002), 1.15; 0.021), leukocytes 1.67; 0.015). For psychopathological variables, i.e., symptomatology, including variables performed not better than null model. A combination hormonal inflammatory markers, well discriminate without disorder. Based on this dataset, however, complex detect non-linear associations markers. These findings need replication future research will reveal respective biomarkers are useful for longitudinal prediction or treatment response.

Background: Increasing evidence points at an important physiological role of the timekeeping system, known as circadian clock (CC), regulating not only our sleep–awake rhythm but additionally many other cellular processes in peripheral tissues. It was shown various cell types that environmental stressors, including ultraviolet B radiation (UV-B), modulate expression genes regulate CC (CCGs) and these CCGs susceptibility for UV-B-induced damage. aim this pilot study to gain further insights into CCs’ putative photocarcinogenesis skin cancer. Methods: Applying RT-PCR, we analyzed two core (brain muscle ARNT-like 1 (Bmal1) Period-2 (Per2)) over several time (0–60 h) HaCaT cells with without 1,25-dihydroxyvitamin D (D3) and/or UV-B conducted a cosinor analysis evaluate effects those conditions on extended mixed-effects linear modeling account both fixed experimental random inter-individual variability. Next, investigated keratinocytes representing different stages photocarcinogenesis, comparing normal (Normal Human Epidermal Keratinocytes—NHEK; p53 wild type), precancerous (HaCaT keratinocytes; mutated status), malignant (Squamous Cell Carcinoma SCL-1; null status) after 12 h under same conditions. Results: We demonstrated cells, Bmal1 showed robust rhythm, while Per2 limited. Overall genes, especially Bmal1, increased following treatment, suppressed overall D3. Both UVB 1,25(OH)2D3 suggested significant phase shift (p < 0.05 acrophase), no specific effect amplitude could be evidenced. Differential BMAL1 were found when compared treatment modalities (UV-B D3) or (NHEK, HaCaT, SCL-1 cells). Conclusions: Comparing epidermal provide independently operating system human skin, which is regulated by disturbed during photocarcinogenesis. Our finding pattern differentially altered UV-B, D3, does support hypothesis may via synthesis vitamin might introducing novel photoprotective property through clock.

Abstract Recent advancements in methodologies such as ecological momentary assessment (EMA) and ambulatory physiology devices have enhanced our ability to measure emotions experienced daily life. Despite the feasibility of EMA for assessing children's youth's emotional self‐reports, combining it with physiological measurements a real‐life context has yet be established. Our scoping review evaluates usability implementing children youth. Due complexities data, this also synthesized existing methodological statistical practices studies. Following PRISMA‐ScR guidelines, we searched screened PsycINFO, PubMed, Web Science electronic databases studies that assessed subjective cardiac or electrodermal responses outside laboratory. initial search resulted 4174 studies, 13 which were included review. Findings showed significant variability EMA, device wear‐time averaging 58.77% study periods data loss due quality issues ranging from 0.2% 77% across signals. Compliance was approximately 60% when combined EMA. The points lack standardized procedures suggests need guidelines designing, processing, analyzing collected contexts. We offer recommendations enhance participant engagement develop standard employing youth emotion, developmental, psychophysiology researchers.

Abstract This chapter addresses autonomic nervous system (ANS) function in nonsuicidal self-injury (NSSI) from the perspective of Research Domain Criteria (RDoC). Based on RDoC framework, ANS, as a major stress-regulatory humans, is primarily considered to be implicated domain arousal/regulatory systems, while it shows associations also other domains. Drawing recent theoretical models providing framework for understanding neurobiology NSSI, suggested that NSSI may result failures physiological and socioemotional stress regulation. In present chapter, after introducing ANS under outlining potential rationale an involvement central mechanisms concepts will presented help understand how typically context psychophysiological research studies. Further, overview indices are frequently derived lab quantify provided. The then reviews existing studies where respective have been measured study different temporal angles (i.e., resting state, response stress, well considering its diurnal rhythmicity). On basis evidence reviewed, discusses current state field concludes with short summary outlook highlighting avenues future research.
