ABSTRACT Vertebrate photoreceptors detect light through a large cilium-based outer segment, which is filled with photopigment-laden membranous discs. Surrounding the base of segment are microvilli-like calyceal processes (CPs). Although CP disruption has been associated altered morphology and photoreceptor degeneration, role CPs remains elusive. Here, we used zebrafish as model to characterize CPs. We quantified parameters report strong disparity in coverage between subtypes. length stable across dark conditions, yet heat-shock inducible expression tagged actin revealed rapid turnover core. Partitioning of gene expression among zebrafish photoreceptor subtypes DOI Creative Commons

Abstract Vertebrate photoreceptors are categorized into two broad classes, rods and cones, responsible for dim- bright-light vision, respectively. While many molecular features that distinguish cones known, gene expression differences among cone subtypes remain poorly understood. Teleost fishes renowned the diversity of their photoreceptor systems. Here, we used single-cell RNA-seq to profile adult in zebrafish, a teleost. We found addition four canonical zebrafish types, there exist subpopulations green red (previously shown be located ventral retina) express red-shifted opsin paralogs ( opn1mw4 or opn1lw1 ) as well unique combination phototransduction genes. Biallelic mutations in the SACS gene, encoding sacsin, cause early-onset autosomal recessive spastic ataxia of Charlevoix-Saguenay (ARSACS), a neurodegenerative disease also characterized by unique and poorly understood retinal abnormalities. While two murine models replicate phenotypic neuronal features observed patients, no phenotype has been described so far. In zebrafish knock-out strain that faithfully mirrors main aspects ARSACS, we impaired visual function due to photoreceptor degeneration, likely caused cell cycle defects progenitor cells. RNA-seq analysis embryos revealed dysfunction proteins related fat-soluble vitamins (e.g., TTPA, RDH5, VKORC) suggested key role neuroinflammation driving defects. Liquid crystal monomers (LCMs) are the raw material for liquid displays, and their use is steadily increasing in electronic products. Recently, LCMs have been reported to be novel endocrine disrupting chemicals, however, mechanisms underlying potential thyroid hormone disruption visual toxicity not well understood. In this study, six widely used fluorinated (FLCMs) were selected determine putative FLCM-induced zebrafish systems. Exposure FLCMs caused damage retinal structures reduced cell density of ganglion layer, inner nuclear photoreceptor layer approximately 12.6-46.1%. also disrupted levels perturbed hypothalamic-pituitary-thyroid axis by affecting key enzymes protein larvae. A hormone-dependent GH3 viability assay supported hypothesis that act as chemicals. It was determined containing aliphatic ring may a higher T3 antagonism compared without an ring. The widespread presence of mercury, a heavy metal found in the environment and used numerous industries domestic, raises concerns about its potential impact on human health. Nevertheless, adverse effects this environmental toxicant at low concentrations are often underestimated. There emerging studies showing that accumulation mercury eye may contribute to visual impairment comorbidity between autism spectrum disorders (ASD) trait impairment. However, underlying mechanism humans rodents is challenging. In response issue, zebrafish larvae with cone-dominated retinal system were exposed 100 nM chloride (HgCl2), according our previous study, followed by light-dark stimulation, social assay, color preference examine functionality relation ASD-like behavior. The presence of contaminants in the environment has increased recent years, and studies have demonstrated that these ability to penetrate blood–retinal barrier directly affect visual systems organisms. Zebrafish are recognized as an ideal model for human eye diseases due their anatomical functional similarities eye, making them efficient versatile organism studying ocular toxicity caused by environmental field toxicology.
