Patients with lateral lymph node metastasis (LLNM) may experience higher locoregional recurrence rates and poorer prognoses compared to those without LLNM, highlighting the need for effective preoperative stratification reliably assess risk LLNM. Recent research has revealed the importance of miRNAs in diagnosis and clinical evolution papillary thyroid cancer (PTC). We aim to identify a specific miRNA profile that could differentiate between subtypes PTC. In this cross-sectional study, total RNA was extracted from paraffin-embedded tissues 43 patients, 17 with an infiltrative follicular variant PTC (iFVPTC) 26 conventional (cPTC). Nine were evaluated using qRT-PCR technology assays. found patterns for cPTC iFVPTC, such as altered both types tumours (miR-146b-5p, miR-181a-5p, miR-221-3p, miR-21-5p miR-222-3p) two significantly expressed only (miR-20b-5p, miR-21-5p). The iFVPTC group presented strong moderate correlations expression data. miR-195-5p, miR-181-5p, miR-146b-5p miR-222 correlated age, tumour size (TS) or lymph node metastases (N), while miR-20b-5p, miR-195-5p miR-181-5p TS, N age group. present study allowed identification signature confirm differences histological TC. Thyroid cancer has emerged as the most rapidly proliferating solid neoplasm. In this study, we included a cohort of patients who underwent sonographic assessment and surgical intervention at Sir Run Shaw Hospital, associated with School Medicine Zhejiang University, spanning from January 2019 to June 2020. Stratification cases was based on combination preoperative ultrasonographic evaluations postoperative histopathological diagnoses, resulting in three distinct groups: high-risk papillary thyroid carcinoma (PTC) labeled C1, low-risk PTC designated C2, control group (N) composed benign tissue adjacent carcinoma. Proteomic phosphoproteomic analyses were conducted specimens. The comparative revealed that proteins up-regulated C1/N C2/N groups predominantly involved functions such amino acid binding, binding phosphorylated compounds, serine protease activity. Notably, like NADH dehydrogenase, ATP synthase, oxidoreductases, iron ion channels significantly elevated C1 versus C2 group. Through meticulous analysis differential expression multiples, statistical significance, involvement metabolic pathways, study identified eight potential biomarkers pertinent metastasis diagnostics, encompassing myosin 10, proline-directed protein kinase, leucine tRNA synthetase, 2-oxo-isovalerate succinic semialdehyde ADP/ATPtranslocase, pyruvate carboxylase, fibrinogen. Therapeutic assays employing metformin, an AMP-activated kinase (AMPK) activator, alongside phosphorylation-specific inhibitor ML-7 targeting Myosin10, demonstrated attenuated cellular proliferation, migration, invasion capabilities cells, accompanied by reduction pools.
