Odorant receptors (ORs) are important class of proteins involved in olfactory behaviour insects. These GPCR-like heptahelical transmembrane with inverted topology compared to GPCR and require a co-receptor (ORco) for their function. OR function can be modulated through small molecules negative modulation beneficial case disease vectors like Aedes aegypti. In this study, we have attempted model the full-length structure ORco due lack experimental structure. Further, screened library natural compounds (>0.3 million) along known repellent against OR4. Many compounds, including those from plants Ocimum tenuiflorum (Holy Basil) Piper nigrum (Black pepper), were found better binding affinity towards repellents DEET providing an alternative existing molecules. Olfaction is a complex recognition process that critical for chemical communication in insects. Though some insect species are capable of discrimination between compounds structurally similar, little understood about how this high level arises. Some insects rely on discriminating enantiomers compound, demonstrating an ability highly selective recognition. The role two major peripheral olfactory proteins olfaction, i.e., odorant-binding (OBPs) and odorant receptors (ORs) has been extensively studied. OBPs ORs have variable capabilities, with found to display specialized binding capability, whilst others exhibit promiscuous activity. A deeper understanding odorant-protein interactions induce response relies further analysis such as structural studies. In review, we explore the potential specific recognition, specifically enantiomeric discrimination. We summarize state research into OBP OR function focus reported examples literature clear by these proteins.

The olfactory sensory neurons of vinegar flies and mice tend to express a single ligand-specific receptor. While this ′one neuron-one receptor′ motif has long been expected apply broadly across insects, recent evidence suggests it may not extend mosquitoes. We sequenced analyzed the transcriptomes 46,000 from antennae dengue mosquito Aedes aegypti resolve all olfactory, thermosensory, hygrosensory neuron subtypes identify receptors expressed therein. find that half coexpress multiple receptors. However, coexpression occurs almost exclusively among genes same family—among odorant (ORs) or ionotropic (IRs). Coexpression ORs with IRs is exceedingly rare. Many coexpressed are duplicates. In other cases, recruitment co-option by placed these together in cells distant paralogs. Close examination data Drosophila reveal rare cases both phenomena, indicating systems two species fundamentally different, but instead fall at different locations along continuum likely encompass diverse insects.

Insects detect odors via a large variety of odorant receptors (ORs) expressed in olfactory sensory neurons (OSNs). The insect OR is heteromeric complex composed ligand-specific receptor and the co-receptor (ORco). In this study, we identified ORco gene cockroach, Periplaneta americana (PameORco), performed RNAi-based functional analysis PameORco. All OSNs basiconic sensilla PameORco received including sex pheromones. trichoid sensilla, PameORco-positive OSN was consistently paired with PameORco-negative tuned to acids. adult cockroaches injected dsRNA at nymphal stage, expression PameORco, odor receptions ORs, its central processing were strongly suppressed. These results provide new insights into molecular basis reception cockroach. long-lasting irreversible effects RNAi would be an effective method for controlling household pest.

Since antiquity, the sense of smell (olfaction) is considered as a distance sense, just like sight and hearing. Conversely, taste (gustation) thought to operate by direct contact, similarly touch. With progress natural sciences, information at molecular, anatomical, neurobiological levels has also contributed taste-smell dichotomy, but much evidence inconsistent with sharp differentiation these two senses emerged, especially when considering species other than humans. In spite this, conflicting been interpreted so that it could conform traditional differentiation. As result, confirmation bias currently affecting scientific research on chemosensory systems hindering development satisfactory narrative evolution chemical communication across taxa. From this perspective, dichotomy loses its validity usefulness. We thus propose unification all modalities into single moving toward synthetic, complex, interconnected perspective gradual processes which vast variety chemicals have become signals are crucially important among within cells, organs, organisms in wide environmental conditions.

Division of labor (DOL) is a characteristic trait insect societies, where tasks are generally performed by specialized individuals. Inside workers focus on brood or nest care, while others take risks foraging outside. Abstract Molecular studies on odorant receptors (ORs), odorant‐binding proteins (OBPs) and the functioning of receptor pheromone signal transduction in fruit fly Bactrocera species have expanded exponentially during past few decades. OBPs contribute to sensing olfactory system (OS) via odorants through sensillum lymph. However, ORs, a family G‐protein‐coupled various other species, exhibit heightened responsiveness multiple chemical odours such as hormones, sensory stimuli neurotransmitters. The apparent mechanism involves combinatorial code encompassing both peripheral antennal lobe processing, facilitating reception sexual pheromones environmental cues. OS is specifically designed recognize process information from volatile signals, these signals play an important function flies. Insects rely chemicals navigate comprehend their surroundings. A mature insect composed two pairs sensillae‐covered palps, antennae primary appendages anterior head. It has been shown that chemosensory gene families odour perception.
