Endothelial mechanics control vascular reactivity and are regulated by the mineralocorticoid receptor (MR) its downstream target, epithelial Na+ channel (ENaC). dysfunction is a hallmark of chronic kidney disease (CKD), but mechanisms poorly understood. We hypothesized that CKD disrupts endothelial in an MR/ENaC-dependent process.Primary human cells were cultured with uremic serum derived from children stage 3-5 (predialysis) or adult hemodialysis (HD) patients healthy controls. The height stiffness glycocalyx (eGC) cortex monitored atomic force microscopy (AFM) using ultrasensitive mechanical nanosensor. In stage-dependent manner, sera induced significant increase eGC incremental reduction height. AFM measurements significantly associated individual pulse wave velocity concentrations gut-derived toxins. Serum HD increased MR expression cortex, effect reversed ENaC antagonists, decreased eNOS NO bioavailability, augmented monocyte adhesion. These data indicate progressive structural damage surface diminishing function identify as mediator CKD-induced dysfunction.

Chronic kidney disease (CKD) is a global health challenge affecting nearly 700 million people worldwide. In the United States alone, Medicare costs for CKD management has reached USD 80 billion per year. While reversing may be possible in future, current strategies aim to slow its progression. For most part, have focused on employing Renin Angiotensin Aldosterone (RAS) inhibitors and optimizing blood pressure diabetes mellitus control. Emerging data are showing that disruption of gut-kidney axis significant impact delaying Recent investigations documented promising results using microbiota-based interventions better manage CKD. This review will summarize evidence explore future possibilities use probiotics, prebiotics, synbiotics, fecal microbial transplant reduce

The uremic toxin indoxyl sulfate (IS) has been related to the development of various medical conditions notably chronic kidney disease (CKD). Hence, quantification this biomarker in biological fluids may be a diagnostic tool evaluate renal system functionality. Numerous analytical methods including liquid chromatography, gas spectroscopy, and electrochemical techniques have since used analyze IS different fluids. current review highlights relevant studies that assessed with special focus on sample preparation, which is essential reduce or eliminate effect endogenous components from matrix bioanalysis.

Purpose of review Cardiovascular disease (CVD) is a significant cause morbidity and mortality in children with chronic kidney (CKD). The CVD CKD multifactorial there are new emerging data regarding prevalence risk factors for this population. Recent findings A number recent publications from longitudinal cohort studies have greatly increased our knowledge about the including hypertension, obesity dyslipidaemia. Masked hypertension isolated nocturnal both correlate surrogate markers children. Obesity adiposity associated an CVD. Markers other than BMI such as waist to height ratio fat-free tissue fat better presence Dyslipidaemia extremely prevalent paediatric population, but lack consensus on treatment. More relationship between bone mineral continue emerge association hyperparathyroidism hypertension. Summary Children multiple potentially modifiable Research focused outcomes needed.

Chronic kidney disease (CKD) is a condition of widespread epidemiology and serious consequences affecting all organs the organism associated with significant mortality. The knowledge on CKD rapidly evolving, especially concerning adults. Recently, more data also appearing regarding in children. itch (CI) common symptom due to various underlying dermatological systemic conditions. CI may appear association termed chronic disease-associated (CKD-aI). CKD-aI relatively well-described literature adults, yet it affects Unfortunately, paediatric particularly scarce. This narrative review aims describe aspects an emphasis children, based available this population extrapolated from Its pathogenesis described details, focusing growing role uraemic toxins (UTs), as well immune dysfunction, altered opioid transmission, infectious agents, xerosis, neuropathy dialysis-associated aspects. Moreover, epidemiological clinical are reviewed few whereas treatment recommendations proposed well, adults own experience managing
