Abstract The muroid rodent Nannospalax galili (syn. Spalax ) is adapted to life in underground burrows and tolerates acute exposure severe hypoxia. Adaptation hypoxia correlated with delayed onset of ageing resistance against tumour formation. becomes five seven times older than its relatives, the mouse rat, without displaying signs or developing ageing-related disorders like cancer. Investigating understanding genes gene regulatory networks might pinpoint novel strategies maintain an extended healthy phenotype humans. Here we analysed compared RNA-Seq data liver, kidney spleen rat subjected 6% O 2 normoxia. We identified differentially expressed pathways common multiple organs rat. Body-wide differences between affected biological processes cell death, defence reactive oxygen species (ROS), DNA repair, energy metabolism, immune response angiogenesis, which altogether play a crucial role ’s adaptation under deprivation. In all organs, mRNA expression associated genome stability maintenance repair was elevated accompanied by lower aerobic metabolism proinflammatory processes. These transcriptomic changes be accountable for extraordinary lifespan cancer resistance. Our results reveal that become candidates investigation molecular bases underlie complex .

In the subterranean rodent (Nanno)spalax galili, evolutionary adaptation to hypoxia is correlated with longevity and tumor resistance. Adapted gene-regulatory networks of Spalax might pinpoint strategies maintain health in humans. Comparing liver, kidney spleen transcriptome data from rat at normoxia, we identified differentially expressed gene pathways common multiple organs both species. Body-wide interspecies differences affected processes like cell death, antioxidant defense, DNA repair, energy metabolism, immune response angiogenesis, which may play a crucial role Spalax's environmental hypoxia. all organs, transcription genes for genome stability maintenance repair was elevated versus rat, accompanied by lower expression aerobic metabolism proinflammatory genes. It is well known that extreme heat events happen frequently due to climate change. However, studies examining the direct health impacts of increased temperature and waves are lacking. Previous reports revealed heatstroke induced acute lung injury pulmonary dysfunction. This study aimed investigate whether exposure fibrosis explore underlying mechanisms. Male C57BL/6 mice were exposed an ambient 39.5 ± 0.5 °C until their core reached maximum or exhaustion state. Lung was observed in lungs heat-exposed mice, with extensive collagen deposition elevated expression molecules, including transforming growth factor-β1 (TGF-β1) Fibronectin (Fn1) (p < 0.05). Moreover, epithelial–mesenchymal transition (EMT) occurred response exposure, evidenced by E-cadherin, epithelial marker, which downregulated, whereas markers EMT, such as connective tissue factor (CTGF) zinc finger transcriptional repressor protein Slug, upregulated tissues Subsequently, cell senescence examination levels both senescence-associated β-galactosidase (SA-β-gal) staining cycle kinase inhibitor p21 significantly Mechanistically, cGAS–STING signaling pathway evoked DNA damage activated In summary, we reported a new finding contributed development early fibrosis-like changes through damage-activated followed cellular senescence.

Heat stress-induced biochemical alterations in ovarian follicles compromise the function of granulosa cells (GCs) and developmental competence oocytes. Summer heat stress can have a far-reaching negative impact on overall fertility reproductive success. Together with stress, rise assisted technologies (ART), potential confounding hazards vitro handling absence systemic body support ART makes it imperative to study ameliorative effects vitamin C under conditions. Using treatment 43 °C for two hours bovine GCs, we studied cell growth, oxidative apoptosis cycle progression together comprehensive metabolomics profiling. Introduction: Acute onset of severe psychiatric symptoms or regression may occur in children with premorbid neurodevelopmental disorders, although typically developing can also be affected. Infections other stressors are likely triggers. The underlying causes unclear, but a current hypothesis suggests the convergence genes that influence neuronal and immunological function. We previously identified 11 pediatric acute-onset neuropsychiatric syndrome (PANS), which two classes related to either synaptic function immune system were found. Among latter, three affect DNA damage response (DDR): PPM1D, CHK2, RAG1. now report an additional 17 cases mutations PPM1D DDR patients acute and/or their clinicians classified as PANS another inflammatory brain condition. Methods: analyzed genetic findings obtained from parents carried out whole-exome sequencing on total cases, included 3 sibling pairs family 4 affected children. Results: include clusters affecting p53 repair (PPM1D, ATM, ATR, 53BP1, RMRP), Fanconi Anemia Complex (FANCE, SLX4/FANCP, FANCA, FANCI, FANCC). hypothesize defects genes, context infection stressors, could contribute decompensated states through increase genomic instability concomitant accumulation cytosolic cells triggering sensors, such cGAS-STING AIM2 inflammasomes, well central deficits neuroplasticity. In addition, increased senescence defective apoptosis responses playing role. Conclusion: These compelling preliminary motivate further functional characterization downstream impact point novel treatment strategies.

Planetary health embraces the concept that long-term human welfare depends on well-being of its ecological systems. Current practices, however, have often ignored this and led to an anthropocentric world, with consequence increased greenhouse gas emissions, heat stress, lack clean water pollution, are threatening environment as well life Homo sapiens many other species. One environmental stressors has been stimulation inflammatory oxidative stress may not only promote common lifestyle diseases, but ageing process.
