Abstract Introduction Enzootic nasal adenocarcinoma (ENA) is a cancer that occurs in goats and sheep infected by enzootic tumour virus. Pathologic examinations are useful for distinguishing tumours from inflammatory hyperplasia. The aim of this study was to describe the pathological characteristics ENA. Material Methods Caprine samples were collected examination. tissue sections stained with haematoxylin-eosin periodic acid-Schiff (PAS) processed immunohistochemical staining. Tumour also routine transmission electron microscopy (TEM). Results histopathological structure exhibited both papillary formations superficial regions tubular or acinar deeper layers, representing distinct structural patterns within same adenocarcinoma. cells positive PAS, mitotic figures rare. Low-differentiated nests epithelial–mesenchymal transition phenomena observed. Immunohistochemical analysis showed strongly pancytokeratin cytokeratin (CK)18, moderately CK7, did not express olfactory marker protein. Kiel 67 labelling index approximately 23%. Retrovirus-like particles distributed inside outside TEM. Conclusion origin site ENA epithelium glandular tubules. This low-grade malignant potential. Cytokeratin 7 CK18 can be considered immunophenotypes identifying cells.

The nonstructural protein 13 (NSP13) of PDCoV is a highly conservative helicase and plays key roles in viral replication. NSP13 contains zinc-binding domain (ZBD), helical Stalk domain, beta-barrel 1B core domain. However, the specific functions these domains remain largely unknown. Here, we expressed purified wild-type NSP13WT various mutants with deletions, activities proteins were analyzed using multiple methods. We found that NSP13ΔZBD possessed abilities to hydrolyze ATP unwind double-stranded nucleic acids, but unwinding efficiency was lower than NSP13WT. In contrast, NSP13ΔZBD-Stalk, NSP13Δ1B, NSP13ΔZBD-Stalk-1B all lost their activity, not ATPase activity. These results revealed deletion ZBD impaired activity NSP13, critical for separate duplexes. identification each this study helpful gain an in-depth understanding overall providing theoretical basis development antiviral drugs targeting helicase.

The cabbage whitefly (A. proletella) (Hemiptera: Aleyrodidae) is a major agricultural pest that primarily targets cruciferous crops, such as cabbage, broccoli, and kale, causing extensive damage through feeding honeydew. However, its associated virome has received limited research attention. In this study, we collected whiteflies in Xinjiang Agricultural University (43.80833 N, 87.56778 E, 882.3 m), systematically identified the RNA of A. proletella successfully three novel iflaviruses (Aleyrodes iflavirus 1 (APIV1), Aleyrodes 2 (APIV2) 3 (APIV3)). APIV1-3 all have 5' structural protein region 3' non-structural region. Phylogenetic sequence identity analyses suggest are members family Iflaviridae. Structural modeling using AlphaFold3 revealed conserved core variable outer loop This study provides valuable insights into diversity proletella, establishing foundation for future on virus-host interactions potential biocontrol applications sustainable agriculture.

Abstract Hendra and Nipah viruses (HNVs), zoonotic paramyxoviruses with >50% case fatality rates, cause fatal encephalitis respiratory disease, yet lack approved therapies. Here, nine rhesus‐derived monoclonal antibodies (mAbs) targeting the fusion glycoprotein (F) prefusion conformation are developed. Four mAbs exhibit first‐rate cross‐neutralization against HNVs, two showing synergistic potency when combined attachment (G)‐specific mAbs. Single‐dose administration of confers robust protection lethal virus challenge in hamsters. Structural insights reveal that 8 9 potent adopt a human IGHV4‐59‐like framework protruding CDRH3 loops, forming pushpin‐shaped paratopes stabilize F‐trimer by occupying vulnerable interprotomer cavities. Systematic mutational profiling identifies 14 prefusion‐locking residues within F ectodomain, classified as i) structural linchpins governing fusogenicity or ii) immunodominant hotspots targeted cross‐neutralizing This work delivers promising therapeutic candidates HNVs provides blueprints for rational design vaccines viral machinery.

Apple viruses pose significant threat to global apple production. In this study, HTS technology was used investigate the virome in Czech Republic. Previously reported viruses, including ACLSV, ASPV, ASGV, ApMV, AGCaV, and CCGaV, were confirmed, near-complete genomes assembled. Additionally, two novel ARWV1 ARWV2 identified for first time Phylogenetic analyses showed low genetic variability among isolates, suggesting a possible recent introduction or limited diversification. contrast, isolates displayed distinct clustering coat protein coding region, separating symptomatic asymptomatic samples, indicating potential involvement of determinants symptom expression. Mixed infections prevalent, with multiple molecular variants AGCaV detected within individual along co-infections involving from different families. Recombination analysis frequent recombination events ACLSV often non-apple parental sequences, their cross-host infections. an interspecific event almond ApMV isolate, PNRSV as minor parent. These findings highlight impact agricultural practices on viral evolution host adaptation. This study demonstrates utility powerful tool uncovering diversity, events, evolutionary dynamics.

Abstract Nucleorhabdoviruses, classified into four distinct genera within the family Rhabdoviridae (subfamily Betarhabdovirinae ), are plant-infecting viruses, characterized by nucleus-associated, enveloped, bacilliform virions. Nucleorhabdoviruses possess an unsegmented, single-stranded, negative-sense RNA genome (ca. 12-15.2 kb) that encodes six to eight proteins. Here, exploring large publicly available metatranscriptomics datasets, we report identification and genomic characterization of 47 novel viruses with genetic evolutionary hallmarks nucleorhabdoviruses. These associated 45 host plant species were previously hidden in public database. Our findings significantly broaden known range nucleorhabdoviruses, including first nucleorhabdovirus ferns gammanucleorhabdoviruses linked dicot hosts. Genetic divergence analyses suggest all these likely represent members species. Phylogenetic reconstruction indicates ten related alphanucleorhabdoviruses, 25 betanucleorhabdoviruses, deltanucleorhabdoviruses gammanucleorhabdoviruses. This study constitutes final chapter a trilogy chronicling data mining expedition cryptic diversity plant-associated rhabdoviruses, journey began varicosaviruses continued cytorhabdoviruses. new yield most comprehensive phylogeny nucleorhabdoviruses date, expanding our understanding illuminating phylogenetic relationships dynamics this virus group. Further, work underscores value large-scale sequence advancing world rhabdoviruses.

A botybirnavirus, Alternaria alternata botybirnavirus 1 (AaBRV1), had been identified from strain SD-BZF-19 isolated diseased watermelon leaves in our previous study. In the current study, AaBRV1 was eliminated its host fungus using single hyphal tip culture method combined with high-temperature treatment to obtain AaBRV1-free strain, which named SD-BZF-19-G14. Compared SD-BZF-19-G14, following infection, colony color of changed, and growth rate, dry weight mycelial biomass, sensitivity difenoconazole, fludioxonil, tebuconazole all decreased. However, virulence not significantly different disease index inoculated SD-BZF-19-G14 being 90.83 81.67, respectively. total 1244 differentially expressed genes (DEGs) were a comparative transcriptome analysis between two strains, Relative SD-BZF-19, number upregulated downregulated DEGs 660 584, Notably, Pfam database annotated that related Major Facilitator Superfamily (MFS) Cytochrome P450 (CYP450) 36 28, To knowledge, this is first documentation biological characteristics induced by infection A. alternata.
