Results from recent clinical trials of antibodies that target amyloid-β (Aβ) for Alzheimer's disease have created excitement and been heralded as corroboration the amyloid cascade hypothesis. However, while Aβ may contribute to disease, genetic, clinical, imaging biochemical data suggest a more complex aetiology. Here we review history weaknesses hypothesis in view new evidence obtained anti-amyloid antibodies. These indicate treatments either no or uncertain effect on cognition. Despite importance definition argue point playing minor aetiological role. We also discuss suggesting concerted activity many pathogenic factors propose evolving multi-factor models will better underpin search effective strategies treat disease.

Curcumin (diferuloylmethane), a polyphenol extracted from the plant Curcuma longa, is widely used in Southeast Asia, China and India food preparation for medicinal purposes. Since second half of last century, this traditional medicine has attracted attention scientists multiple disciplines to elucidate its pharmacological properties. Of significant interest curcumin's role treat neurodegenerative diseases including Alzheimer's disease (AD), Parkinson's (PD) malignancy. These all share an inflammatory basis, involving increased cellular reactive oxygen species (ROS) accumulation oxidative damage lipids, nucleic acids proteins. The therapeutic benefits curcumin these appear multifactorial via regulation transcription factors, cytokines enzymes associated with (Nuclear factor kappa beta) NFκB activity. This review describes historical use medicine, chemistry, stability biological activities, anti-cancer, anti-microbial, anti-oxidant, anti-inflammatory further discusses pharmacology provides new perspectives on potential limitations. Especially, focuses detail effectiveness mechanism actions treating such as brain malignancies.

A highly efficient fluorosensor based on ultrathin graphitic carbon nitride (g-C3N4) nanosheets for Cu2+ was developed. In the absence of metal ions, exhibit high fluorescence; strong coordination Lewis basic sites them to however, causes fluorescence quenching via photoinduced electron transfer leading qualitative and semiquantitative detection ions. This exhibits selectivity toward Cu2+. The whole process can be completed within 10 min with a limit as low 0.5 nM. use test paper enables naked-eye 0.1 nmol. practical this sensor determination in real water samples also demonstrated.

Whilst being environmentally abundant, aluminum is not essential for life. On the contrary, a widely recognized neurotoxin that inhibits more than 200 biologically important functions and causes various adverse effects in plants, animals, humans. The relationship between exposure neurodegenerative diseases, including dialysis encephalopathy, amyotrophic lateral sclerosis Parkinsonism dementia Kii Peninsula Guam, Alzheimer's disease (AD) has been suggested. In particular, link subject of scientific debate several decades. However, complex characteristics bioavailability make it difficult to evaluate its toxicity therefore, remains be established. Mounting evidence suggested significance oligomerization β-amyloid protein neurotoxicity molecular mechanism AD pathogenesis. Aluminum may play crucial roles as cross-linker oligomerization. Here, we review detailed based on our own studies recent literatures. Our aim revisit integrate amyloid cascade hypotheses context interactions with other metals.

Dementia risk reduction is a major and growing public health priority. While certain modifiable factors for dementia have been identified, there remains substantial proportion of unexplained risk. There evidence that environmental may explain some this Thus, we present the first comprehensive systematic review dementia. We searched PubMed Web Science databases from their inception to January 2016, bibliographies articles, articles related publically available data. Articles were included if they examined association between an factor dementia. Studies with another outcome (for example, cognition), physiological measure exposure, case studies, animal studies nutrition excluded. Data extracted individual which were, in turn, appraised methodological quality. The strength consistency overall each identified was assessed. We screened 4784 60 review. Risk considered six categories: air quality, toxic heavy metals, other trace elements, occupational-related exposures, miscellaneous factors. Few took life course approach. at least moderate implicating following factors: pollution; aluminium; silicon; selenium; pesticides; vitamin D deficiency; electric magnetic fields.Studies varied widely size quality therefore must be circumspect our conclusions. Nevertheless, extensive suggests future research could focus on short list Furthermore, further robust, longitudinal repeated measures exposures are required confirm these associations.
