The coronavirus disease 2019 (COVID-19) pandemic triggered an unprecedented concentration of economic and research efforts to generate knowledge at unequalled speed on deregulated interferon type I signalling nuclear factor kappa light chain enhancer in B-cells (NF-κB)-driven interleukin (IL)-1β, IL-6, IL-18 secretion causing cytokine storms. translation the how resulting systemic inflammation can lead life-threatening complications into novel treatments vaccine technologies is underway. Summary Adjuvants, as the name indicates, are adjoined material aimed to assist in functioning when added vaccines they meant boost effect and strongly stimulate immune system. The response of system can be unpredictable, autoimmune/inflammatory syndrome induced by adjuvants (ASIA) was developed address possible adverse reactions an autoimmune inflammatory type that may caused adjuvants. While ASIA, a syndrome, coined defined 2011; reports describing patients with vague nonspecific clinical symptoms following vaccinations appeared much earlier. In other words, ASIA came define, arrange, unite variety symptoms, related autoimmunity, not vaccine itself, rather adjuvant part such aluminum, among others. Since the emergence of COVID-19 pandemic at end 2019, a massive vaccination campaign has been undertaken rapidly and worldwide. Like other vaccines, vaccine is not devoid side effects. Typically, adverse effects include transient headache, fever, myalgia. Endocrine organs are also affected by The major SARS-CoV-2 vaccine-associated endocrinopathies reported since beginning thyroid pancreas disorders. vaccine-induced pituitary diseases have become more frequently described in literature. We searched PubMed/MEDLINE for commentaries, case reports, series articles reporting disorders following vaccination. search was reiterated until September 2022, which eight reports were found. In all cases, there no personal or familial history disease described. All patients had previous infection prior to episode. Regarding type vaccines administered, 50% received (BNT162b2; Pfizer–BioNTech) (ChAdOx1 nCov-19; AstraZeneca). five disorder developed after first dose corresponding vaccine. types disorder, hypophysitis (variable clinical aspects ranging from lesion stalk thickness) three apoplexy. time period between ranged one seven days. Depending on each case’s follow-up time, complete remission obtained apoplexy cases but only with (persistence central diabetes insipidus). Both quantity quality published data about inconveniences limited. Pituitary disorders, unlike occur very quickly (less than days versus two months disease). This partially explained ease reaching pituitary, small gland. Therefore, this gland overspread, explains speed onset symptoms (especially ADH deficiency rapid deficit evocative symptoms). SARS-CoV-2 mRNA vaccination can entail chronic fatigue/dysautonomia tentatively termed post-acute COVID-19 syndrome (PACVS). We explored receptor autoantibodies and interleukin-6 (IL-6) as somatic correlates of PACVS. Blood markers determined before six months after first-time healthy controls (N = 89; 71 females; mean/median age: 39/49 years) were compared with corresponding values PACVS-affected persons 191; 159 40/39 exhibiting (≥three symptoms for ≥five the last vaccination) not due to infection and/or confounding diseases/medications. Normal response encompassed decreases in 11 antibodies (by 25–50%, p < 0.0001), increases two 15–25%, 0.0001) normal IL-6. In PACVS, serological vaccination–response appeared significantly (p altered, allowing discrimination from post-vaccination state (sensitivity 90%, by increased Angiotensin II type 1 (cut-off ≤ 10.7 U/mL, ROC-AUC 0.824 ± 0.027), decreased alpha-2B adrenergic ≥ 25.2 0.828 0.025) IL-6 2.3 pg/mL, 0.850 0.022). PACVS is thus indicated a delineated/detectable diagnostic blood markers.

Since vaccines are in fact manufactured chemical compounds such as drugs, the appearance of side effects following their use is not surprising. Similarly, main goal to stimulate immune system bringing out production protective antibodies, autoimmune-related a consequence increased activity do seem irrational. Fortunately, rate low; however, importance reporting adverse events vaccinations, understanding mechanisms behind appearance, making early diagnosis, and appropriate treatment cannot be overemphasized. In fact, vaccines, particularly those based on adjuvants, were reported long before introduction autoimmune/inflammatory syndrome induced by adjuvants (ASIA). Nevertheless, ASIA gathered united under one title, step which helped organize research call for better stimulators than adjuvants. New technologies methods clearly noticed during pandemic COVID-19 after mRNA-based vaccines. our current paper, we introduce notion autoimmune nature, ASIA, linked with including recent The transition from idea work.

Purpose The purpose of this study was to describe the current knowledge on potential endocrine adverse effects post-COVID-19 vaccines. Methods A PubMed/MEDLINE, Web Science, and Scopus research performed. Case reports, case series, original studies, reviews written in English published online up 31 July 2022 were selected reviewed. final reference list defined based relevance each paper scope review. Results available data showed that side are generally rare with favorable outcome, being thyroid disorders most common. Conversely, type 1 diabetes mellitus rare; adrenal pituitary events even anecdotal. Finally, clinical studies suggest no impact female reproductive system male couple fertility. Conclusion Overall, these show that, after 2 years COVID-19 vaccines, is not heavily threatened.

Abstract Background The development of an effective vaccine is a powerful tool to contain the global spread coronavirus disease 2019 (COVID-19). Still, it raises potential safety concerns about subsequent enhancement associated immunopathology. Increasing evidence shows that endocrine system, including hypophysis, may be involved in COVID-19. Moreover, occasional but increasing reports disorders involving thyroid have been reported after severe acute respiratory syndrome 2 (SARS-CoV-2) vaccine. Among them, few cases encompass pituitary. Here we report rare case central diabetes insipidus following SARS-CoV-2 vaccination. Case presentation We 59-year-old female patient with 25-year history Crohn's long-term remission, who presented sudden onset polyuria eight weeks administration mRNA Laboratory evaluation was consistent isolated insipidus. Magnetic resonance imaging displayed involvement infundibulum and posterior hypophysis. Eighteen months vaccination, she still under desmopressin treatment had stable pituitary stalk thickening on magnetic imaging. Although disease-associated hypophysitis has reported, scarce. In absence other recognizable causes hypophysitis, believe hypophysis our triggered by Conclusions potentially Further studies are needed understand better mechanisms underlying autoimmune endocrinopathies context COVID-19 infection

Vaccines against coronavirus disease 2019 (COVID-19) have been distributed in most countries for the prevention of onset and aggravation COVID-19. Recently, there increasing numbers reports on new-onset autoimmune autoinflammatory diseases following COVID-19 vaccination, however, only little information is available long-term safety these vaccines. Here, we experienced three cases rheumatic one case each rheumatoid arthritis (RA), anti-neutrophil cytoplasmic antibody (ANCA)-associated vasculitis (AAV) systemic lupus erythematosus (SLE). The symptom ranged from day to a few days vaccination. patients AAV SLE were treated successfully with glucocorticoid therapy, patient RA died due In literature review which including seven RA, 37 18 SLE, mean time vaccination was approximately 11 12 days. Most improved glucocorticoid, immunosuppressive drugs biologic agents. Although such adverse effects are rare, vaccines useful prevent severity infections, continued accumulation similar important terms examining understanding pathogenic mechanism diseases.
