Keratinocytes play an essential role in the inflammatory phase of wound regeneration. In addition to migrating and proliferating for tissue regeneration, they produce a large amount cytokines that modulate process. Previous studies have shown subthermal treatment with radiofrequency (RF) currents used capacitive resistive electric transfer (CRET) therapy promotes proliferation HaCat keratinocytes modulates their cytokine production. Although physical therapies been anti-inflammatory effects variety experimental models patients, knowledge biological basis these is still limited. The aim this study was investigate effect CRET on keratinocyte proliferation, production (IL-8, MCP-1, RANTES, IL-6, IL-11), TNF-α secretion, expression MMP9, MMP1, NF-κB, ERK1/2, EGFR. Human (HaCat) were treated intermittent 448 kHz current (CRET signal) conditions different periods time. Cell analyzed by XTT assay, ELISA, NF-κB activation immunofluorescence, EGF receptor immunoblot. Compared control, increases transient release TNF-α, IL-6 while decreasing IL-8. addition, it modifies MMPs activates EGFR, ERK1/2 proteins. Our results indicate reasonably through ERK1/2/NF-κB pathway, ultimately modulating response human keratinocytes.
