Three-dimensional bioprinting allows for the fabrication of structures mimicking tissue architecture. This study aimed to develop a gelatin-based bioink bioprinted simplified skin model. The printability and chemical-physical properties were evaluated by varying concentrations gelatin (10, 15, 20%) in semi-crosslinked alginate-based calcium chloride (100, 150, 200 mM) post-printing crosslinking. For increasing concentration, formulations have shear thinning behavior with viscosity, filament bending angle increases, spreading ratio value approaches 1, shape fidelity printing resolution improve. However, formulation containing 20% was not homogeneous, resulting also poor properties. morphology pores, degradation, swelling depend on CaCl2 concentrations, but significant way. samples 15% crosslinked 150 mM been selected bilayer model human fibroblasts keratinocytes. showed homogeneous distribution viable proliferating cells over up 14 days vitro culture. allowed 3D model, potential applications bioactivity pro-reparative molecules drug evaluation.

Abstract Preclinical animal research has long been a cornerstone in evaluating the efficacy, toxicity, and safety of potential drug treatments before they proceed to human clinical trials. However, given intricate nature physiology complexities diseases such as cancer, this paper critically examines role experimentation translational research, both from epistemological ethical viewpoints. We argue that obligation protect animals extends beyond their instrumental value for benefit; it is rooted intrinsic well-being. Consequently, we advocate paradigm shift medical research: adoption new approach methodologies (NAMs) not merely supplementary tools but complete replacements use studies. In context, replacement emerges key principle—an imperative should be prioritized over all other considerations.

Kidney disease encompasses a wide spectrum of conditions, ranging from simple infections to chronic kidney disease. When the kidneys are unable filter blood and remove waste products, these abnormalities can lead failure. In severe cases failure, transplantation is considered only definitive treatment. Worldwide, World Health Organization (WHO) repeatedly emphasizes importance organ donation increasing rates. Many countries implement national programs promote culture improve patient access transplantation. The extent which this procedure performed varies across influenced by several factors, including volume donation, medical infrastructure, technology health policies. However, transplant comes with challenges problems that impact its success. tissue engineering new approach shows promise for repairing replacing damaged tissue. This article reviews recent advances in engineering, focusing on engineered structures such as hydrogels, electrospinning, 3D bioprinting, microfluidic systems. By mimicking extracellular environment kidney, provide suitable conditions growth development cells. role formation vessels, mimicry functions field were also discussed. results study show has high potential treating diseases reducing need achieve clinical application technology, further research required biocompatibility, vascularization long-term performance tissues.
