Mitochondria are of great importance in cell biology since they major sites adenosine triphosphate (ATP) production and widely involved different cellular pathways the response to stress. During ATP production, reactive oxygen species (ROS) can be produced. While a small amount ROS may important for regulation physiological processes, at elevated levels turn into harmful agents leading damage. However, date, there no surveys which address above issues. To fill this gap, valuable collect recent findings about role mitochondria function, not only increase knowledge but also clinical applications. Besides abstinence, no effective treatment exists for alcohol-related liver disease (ALD), a dreaded consequence of alcohol abuse. In this study, we assessed the roles on ALD dual specificity phosphatase-1 (DUSP1), an hepatoprotective enzyme, and Cullin-1 (CUL1), member E3 ubiquitin ligase complex that exerts also transcriptional suppression mitochondrial genes. Alcohol downregulated hepatic DUSP1 expression in wild-type mice. Notably, transgenic (Dusp1Tg ) mice showed resistance to alcohol-mediated dysfunction, as evidenced by decreased AST/ALT activity, improved metabolism, suppressed fibrosis, inflammation, oxidative stress. Functional experiments demonstrated overexpression prevents damage hepatocytes through restoring mitophagy. Accordingly, pharmacological blockade mitophagy abolished actions DUSP1. Molecular assays binds cytosolic CUL1 its translocation nucleus. Importantly, silencing restored transcription p62 Parkin, resulting activation, sustained integrity hepatocyte function upon These results indicate downregulation interrupts DUSP1/CUL1 interaction, leading nuclear inhibition via repression Parkin. Thus, targeting DUSP1/CUL1/p62 axis will be key approach restore well alleviate symptoms ALD.

Percutaneous coronary intervention (PCI), as an essential treatment for artery disease, has significantly improved the prognosis of patients with large lesions. However, some continue to experience myocardial ischemic symptoms post-procedure, largely due microvascular dysfunction (CMD). The pathophysiological mechanisms CMD are complex and involve endothelial dysfunction, remodeling, reperfusion injury, metabolic abnormalities. Moreover, components syndrome, including obesity, hyperglycemia, hypertension, dyslipidemia, exacerbate occurrence progression through multiple pathways. This review systematically summarizes latest research advancements in after PCI, its pathogenesis, diagnostic techniques, management strategies, future directions. For diagnosis, invasive techniques such flow reserve index microcirculatory resistance, well non-invasive imaging modalities (positron emission tomography cardiac magnetic resonance), provide tools early detection. In terms management, a multi-level strategy is emphasized, incorporating lifestyle modifications (diet, exercise, weight control), pharmacotherapy (vasodilators, hypoglycemic agents, statins, modulators), traditional Chinese medicine, specialized treatments (enhanced external counterpulsation, surgery, lipoprotein apheresis). challenges remain treatment, limitations lack personalized strategies. Future should focus on interactions between risks, aiming optimize therapeutic strategies improve long-term post-PCI.
