Abstract In this study, a synthetic approach is reported for generating hepta-branched β-cyclodextrins (CDs) bearing heterogeneous carbohydrate residues at their primary side via one-pot process with simultaneous click chemistry reaction. The reactions were performed by reacting two or three different species of 2-propynylated glycosides hepta-azide functional β-CD various reaction molar ratios. 2-Propynylated acted as sources embedded into β-CD. generated several desired derivatives varying densities the carbohydrates in process. article describes effects combination and ratios reactions.

Osteoarthritis is more prevalent than any other form of arthritis and characterized by the progressive mechanical deterioration joints. Glucosamine, an amino monosaccharide, has been used for over fifty years as a dietary supplement to alleviate osteoarthritis-related discomfort. Silibinin, extracted from milk thistle, modifies degree glycosylation target proteins, making it essential component in treatment various diseases. In this study, we aimed investigate functional roles glucosamine silibinin cartilage homeostasis using TC28a2 cell line. Western blots showed that suppressed N-glycosylation gp130, EGFR, N-cadherin proteins. Furthermore, both differentially decreased increased proteins such Snail, KLF4 cells. We observed compounds dose-dependently induced proliferation Our MitoSOX DCFH-DA dye data 1 µM mitochondrial reactive oxygen species (ROS) generation cytosol ROS generation, whereas JC-1 red aggregates, resulting increase red/green fluorescence intensity ratio, while all tested concentrations green monomers, decreases ratio. increasing subG1 S populations decreasing G1 G2/M with amounts glucosamine, led increases subG1, S, MTT cytotoxicity cells dose-dependent manner. Regarding endoplasmic reticulum stress, expression CHOP level p-eIF2α/eIF2α. With respect O-GlcNAcylation status, reduced levels O-GlcNAc transferase hypoxia-inducible factor alpha. examined mRNAs related these processes. summary, our findings demonstrated modulated cellular proliferation, membrane potential, cycle profile, autophagy. ABSTRACT Glycosylation is a common and complex post‐translational modification (PTM) of proteins, involving the attachment glycans under regulation various enzymes such as glycosyltransferases. facilitates correct folding peptide chains, modifies protein conformation activity, enhances stability influences inter‐protein interactions. N‐glycosylation O‐glycosylation are two prevalent forms, encompassing wide range modifications, including sialylation, fucosylation galactosylation. In skin tumours, abnormal glycosylation promotes tumour cell proliferation, migration, invasion metastasis, anti‐tumour immunity, potentially affects immune checkpoint therapy. inflammatory autoimmune diseases, in T B lymphocyte subpopulations regulates antigen recognition, signal transduction, factor secretion immunoglobulin function, disrupting system homeostasis impacting biologic therapy efficacy. correlates with severity activity serving potential biomarker for diagnosis, condition assessment prognosis determination. This review provides an overview role melanoma, basal carcinoma, squamous psoriasis, systemic lupus erythematosus, dermatomyositis aging. It analyses biosynthetic process glycosylation, elucidates functional changes glycoproteins their metabolism, offers theoretical basis developing new targeted therapies.

Abstract Pelvic organ prolapse (POP) is a common and distressing condition affecting women, particularly those with history of vaginal delivery. The impact extracellular vesicles derived from adipose‐derived mesenchymal stem cells (ADSC‐EVs) on pelvic floor tissue injury remains unclear. Due to their short half‐life rapid clearance in vivo, ADSC‐EVs lose efficacy quickly. To address this, an injectable tetra‐PEG hydrogel encapsulate (PEG@EVs) developed. formed by tetra‐PEG‐NH 2 tetra‐PEG‐NHS through ammonolysis reaction, leading the formation amide bonds within seconds. Vaginal wall POP patients shows disruption matrix, lipid peroxidation, inflammation. In vitro, significantly reduce H₂O₂‐induced oxidative stress, oxidation, apoptosis, while enhancing expression Nrf2 its downstream targets—CAT, NQO1, HO‐1, SOD2. also upregulate GPX4 SLC7A11, reducing mitochondrial damage mitigating ferroptosis. inhibitor ML385 reverses these protective effects. rat model childbirth injury, PEG@EVs treatment promotes nuclear translocation, induces M1‐to‐M2 macrophage conversion, reduces inflammation, stimulates collagen deposition, thereby accelerating repair. findings this study may serve as foundation for early targeted intervention POP, representing promising therapeutic approach.

Specific protein 1 (Sp1) is pivotal in sustaining baseline transcription as well modulating cell signaling pathways and factors activity. Through interactions with various proteins, especially factors, Sp1 controls the expression of target genes, influencing numerous biological processes. Numerous studies have confirmed Sp1’s significant regulatory role pathogenesis cardiovascular disorders. Post-translational modifications (PTMs) Sp1, such phosphorylation, ubiquitination, acetylation, glycosylation, SUMOylation, S-sulfhydration, can enhance or modify its transcriptional activity DNA-binding stability. These also regulate across different types. crucial regulating non-coding gene proteins response to pathophysiological stimuli. Understanding PTMs advances our knowledge controlling stability during disease onset progression. Abstract Necrotizing enterocolitis (NEC) is a life-threatening condition affecting preterm infants, sometimes necessitating surgical treatment. This study aimed to analyze differentially expressed proteins (DEPs) and access their biological clinical significance in the plasma of neonates with NEC. Peripheral blood samples were collected from NEC infants at various time points, was separated. Data-independent acquisition (DIA) technology utilized identify DEPs among patients different stages. Bioinformatic analyses, including Gene Ontology Kyoto Encyclopedia Genes Genomes, protein-to-protein interaction analyses performed on DEPs. External datasets, along receiver operating characteristic curves gene set enrichment analysis, used clinically biologically validate findings. between pre-NEC groups indicated reduced protein, heme, nitrogen, purine nucleotide biosynthesis during formation. In addition, enriched points suggested reconstructed extracellular matrix, aberrant B-lymphocyte immune responses, decreased glycosaminoglycan levels progression. These findings both validated using external datasets. Our highlights relevance proteomics patients. demonstrates key pathways involved pathogenesis establishes DIA mass spectrometry as powerful noninvasive tool for evaluating predicting formation Graphical
