Alzheimer's disease (AD) is characterized by oxidative stress-mediated memory dysfunction and neuronal cell death. This study investigated the effects of an ethanol extract from Bauhinia coccinea (EEBC) on impairment damage in a deficit mouse model. EEBC was administered to ICR mice at doses 50, 100, or 200 mg/kg daily for 3 weeks. Cognitive induced via scopolamine (SCO) injection. Brain tissues were analyzed acetylcholine (ACh) levels, acetylcholinesterase (AChE) activity, apoptosis, antioxidant markers. Behavioral tests showed that SCO injection loss, whereas significantly ameliorated SCO-mediated impairment. regulated cholinergic system decreasing ACh levels enhancing AChE activity. Nissl staining immunohistochemistry NeuN exerted neuroprotective SCO-injected brains. Moreover, reduced number terminal deoxynucleotidyl transferase dUTP nick end labeling-positive apoptotic cells increased treatment. also reversed SCO-induced changes apoptosis-related protein expression brain tissues. Furthermore, malondialdehyde activated catalase SCO-administered Quantitative RNA sequencing involvement lipid metabolism function regulation. Thus, promising candidate attenuating AD progression as it targets apoptosis.

Abstract Alzheimer’s disease (AD) is a widespread condition that affects adults and the community considerably. The causes are yet unknown, except from advanced age genetic predisposition. Natural products provided advantageous advantages for managing AD due to their efficacy, safety, accessibility. memory boosting effects of chemically characterized Ipomoea carnea ethanol extract (IPC-EtOH) on behavioral, biochemical, histological, molecular levels against cognitive impairment induced by AlCl 3 exposure in rats were assessed using donepezil as reference drug. Behavioral tests (spontaneous alternation T-maze open field test) assays GSK3β, CREB, FOXO1a, β-secretase, tau, oxidative stress biomarkers, histopathology, immunohistochemistry cyclooxygenase 2 (COX-2) conducted. chemical profiling IPC-EtOH UPLC-ESI–qTOF-MS coupled with networking revealed identification 83 bioactive metabolites, including pyrrolizidine alkaloids cinnamic acid derivatives which previously undescribed this species. injection significantly elevated β -secretase, GSSG, GSK-3 , FOXO3a down regulated CAT, SOD, strong COX-2 immunoexpression cortex hippocampus compared controls. Oral co-administration or -treated restored near-normal function these brain regions, attenuating spatial learning, memory, locomotor impairments. These results suggest could be promising therapy mitigating aluminum-induced neurotoxicity, though further studies needed elucidate its precise mechanisms action. outcomes emphasize I. extract’s potential an appealing demonstrating neuroprotective memory-enhancing properties having -induced impairment.

Cognitive impairment (COI) is a prevalent complication across spectrum of brain disorders, underpinned by intricate mechanisms yet to be fully elucidated. Neurons, the principal cell population nervous system, orchestrate cognitive processes and govern balance. Extensive inquiry has spotlighted involvement Foxo3a in COI. The regulatory cascade transactivation implicates multiple downstream signaling pathways encompassing mitochondrial function, oxidative stress, autophagy, apoptosis, collectively affecting neuronal activity. Notably, expression activity profile are subject modulation via various modalities, including methylation promoter, phosphorylation acetylation protein. Furthermore, upstream such as PI3K/AKT, SIRT family, diverse micro-RNAs intricately interface with Foxo3a, engendering alterations function. Through several routes, regulates dynamics, thereby modulating onset or amelioration COI Alzheimer’s disease, stroke, ischemic injury, Parkinson’s traumatic injury. potential therapeutic target, clinical drugs small molecules have been preliminarily shown cognitive-enhancing effects that indirectly affect Foxo3a. Particularly noteworthy randomized, controlled, placebo trials illustrating significant enhancement achievable through autophagy modulation. Here, we discussed role neuron-mediated common cognitively impaired diseases.
