The quality of oocytes determines the development potential an embryo and is dependent on their timely fertilization after ovulation. Postovulatory oocyte aging inevitable factor during some assisted reproduction technology procedures, which results in poor rates impairs development. We found that fisetin, a bioactive flavonol contained fruits vegetables, delayed postovulatory mice. Fisetin improved aged inhibited Sirt1 reduction oocytes. increased GSH level Sod2 transcription to inhibit ROS accumulation Meanwhile, fisetin attenuated aging-induced spindle abnormalities, mitochondrial dysfunction, apoptosis. At molecular level, decreased aberrant expression H3K9me3. In addition, levels Tfam genes Co2 Atp8 by upregulating Finally, inhibition reversed anti-aging effects fisetin. Taken together, Sirt1.

Spermidine (SPD) is an intermediate compound in the polyamine metabolism which takes critical part a variety of cellular processes. In particular, it has been reported to exert anti-aging effects, suppress age-related diseases, and extend lifespan across species. However, whether favorable influence on quality postovulatory aged oocytes remains elusive.

Postovulatory aging is known to impair the oocyte quality and embryo development due oxidative stress in many different animal models, which reduces success rate or pregnancy human assisted reproductive technology (ART) livestock timed artificial insemination (TAI), respectively. Salidroside (SAL), a phenylpropanoid glycoside, has been shown exert antioxidant antitumor effects. This study aimed investigate whether SAL supplementation could delay postovulatory process by alleviating stress. Here, we show that decreases malformation recovers mitochondrial dysfunction including distribution, membrane potential (ΔΨ) ATP content aged oocytes. In addition, treatment alleviates aging-caused such as higher reactive oxygen species (ROS) level, lower glutathione (GSH) reduced expression of antioxidant-related genes. Moreover, cytoplasmic calcium ([Ca2+]c) ([Ca2+]mt) SAL-treated oocytes return normal levels. Notably, suppresses aging-induced DNA damage, early apoptosis improves spindle assembly oocytes, ultimately elevating developmental rates quality. Finally, RNA-seq confirmatory experience showed promotes protective autophagy activating MAPK pathway. Taken together, our research suggests supplementing an effective feasible method for preventing preserving quality, potentially contributes improving successful ART TAI.

Decrease in quality of postovulatory aged oocytes occurs due to oxidative stress and leads low fertilization development competence. It is one the main causes that exerting detrimental effect on success rate assisted reproductive technology (ART). Auraptene (AUR), a citrus coumarin, has been reported possess an antioxidant effects other tissues. In this study, we aimed confirm potential AUR delay oocyte aging process by alleviating stress. Superovulated mouse metaphase second meiosis (MII) were exposed 0, 1 or 10 μM for 12 h vitro aging. addition culture medium recovered abnormal spindle chromosome morphology mitigated mitochondrial distribution membrane (ΔΨ) oocytes. AUR-treated also showed suppressed stress, with lower reactive oxygen species (ROS) levels, higher glutathione (GSH) levels increased expression several genes involved antioxidation. Furthermore, significantly elevated embryo developmental rates. Oocytes exhibited morphokinetics very similar those control group. Altogether, these data allowed us conclude improved suggest as effective clinical supplement candidate prevent

Abstract Ovulation, fertilization, and embryo development are orchestrated synchronized processes essential for the optimal health of offspring. Postovulatory aging disrupts this synchronization impairs oocyte quality. In addition, causes fertilization loss poor development. This investigation aimed to unravel endpoint in vitro common carp (Cyprinus carpio) understand involvement apoptosis postovulatory death. It was observed that ability significantly declined (P < 0.001) at 8-h poststripping (HPS), subsequently triggering advanced stage aging, i.e., 48 HPS. process included an increase proapoptotic transcripts (fas, bax, cathepsin D, caspase 8, 9, 3a) 0.05), elevated levels 3 protein activation enzyme 0.001), a key player apoptosis, oocytes. Oocyte quality is crucial for successful fertilization and subsequent embryonic development. Post-ovulatory aging leads to reduced oocyte impaired embryogenesis, representing an unavoidable challenge in terms of certain assisted reproductive techniques. Diosmetin (DIOS), a natural flavonoid found lemons, spearmint, spider moss, exhibits antioxidant, anti-inflammatory, anti-apoptotic properties. However, its effects on the mature porcine oocytes vitro remain unexplored. This study investigated impact DIOS aging. In IVM medium, fresh were cultured 44 h, while 68 h. Following addition varying concentrations (0.01, 0.1, 1 μM) DIOS-treated group was The results demonstrated that 0.1 μM significantly improved blastocyst rates cell counts, reactive oxygen species (ROS), elevated glutathione (GSH) levels, enhanced mitochondrial function, decreased markers autophagy (LC3B), apoptosis (annexin V), endoplasmic reticulum stress (CHOP), senescence (SA-β-Gal). Furthermore, treatment upregulated expression relevant genes compared aged group. These findings suggest effectively delays

The ovaries play a crucial role in both the endocrine health and fertility of adult women. fundamental functional units ovaries, primordial follicles, form during fetal development constitute ovarian reserve. Ovaries age prematurely comparison to other organs, with quality oocytes declining steeply prior entire reserve becoming depleted, usually around 50. Despite pivotal women's overall health, surprisingly little is known about mechanisms controlling follicle dormancy, growth activation, atresia, maturation, oocyte quality. Understanding function on cellular molecular level increasingly important for several reasons. First, global trend women delaying childbirth creates growing population patients wishing conceive when quantity their are already critically low. Second, conditions affecting such as polycystic ovary syndrome endometriosis, widespread, yet diagnosis treatment still present challenges.
