Circular ribonucleic acid (RNA) protein kinase C iota (circ-PRKCI, hsa_circRNA_0122683) has been previously reported to be involved in the development of sepsis. However, knowledge regarding potential role and mechanism circ-PRKCI sepsis-induced acute lung injury (ALI) is unclear. An vitro cellular model sepsis-ALI was simulated by treatment lipopolysaccharide (LPS) human pulmonary alveolar epithelial cells (HPAEpiCs). The expression plasma samples from sepsis patients with or without ALI as well cell determined quantitative real-time PCR (qRT-PCR). diagnostic utility analyzed using receiver operating characteristic (ROC) curves. levels iron content (Fe2+), glutathione (GSH), malondialdehyde (MDA), reactive oxygen species (ROS) were detected corresponding commercial kits. assessment viability production pro-infammatory cytokines (IL-6, IL-1β TNF-α) measured Cell Counting Kit-8 (CCK-8) enzyme-linked immunosorbent assay (ELISA). targeting relationship between miR-382-5p predicted bioinformatics analysis, subsequently confirmed luciferase reporter RNA immunoprecipitation (RIP) assays. Results shows that decreased but increased observed model. area under curve values ROC curves for differentiating septic healthy individuals non-ALI 0.996 0.999, respectively. Functional assays revealed enforced alleviated LPS-induced ferroptosis inflammatory response HPAEpiCs, which reversed Erastin FIN56 administration. Mechanistically, identified a sponge negatively regulated expression. Further rescue experiments showed overexpression could compromise anti-ferroptosis anti-inflammatory effects on HPAEpiCs. Our study demonstrated may promising biomarker diagnosis. inhibits sponging miR-382-5p, indicating circ-PRKCI/miR-382-5p axis might novel therapeutic target ALI.
