The submergence tolerance of rice is a key factor in promoting direct seeding technology and resisting flood disasters. Dongxiang wild (DXWR) has strong tolerance, but its genetic basis still unclear. Here, we report quantitative trait loci (QTLs) analysis for hypoxic germination rate (HGR), seedling (HSR), budlet survival (BSSR) (SSSR) using linkage map the backcross recombinant inbred lines (BRILs) that were derived from cross DXWR, an indica cultivar, GZX49. A total 20 QTLs related to detected, explaining phenotypic variations ranging 2% 8.5%. Furthermore, transcriptome sequencing was performed on seeds seedlings DXWR before after submergence. During seed stages, 6306 3226 differentially expressed genes (DEGs) detected respectively. Gene ontology (GO) Kyoto encyclopedia genomes (KEGG) analyses conducted these genes. Using data, combined with qRT-PCR, sequence comparison, bioinformatics, LOC_Os05g32820 putatively identified as candidate gene qHGR5.2 co-located HGR SSSR. These results will provide insights into mechanism improving tolerance.

Seed vigor is a complex trait encompassing seed germination, seedling emergence, growth, longevity, and stress tolerance, all are crucial for direct seeding in rice. Here, we report that the AP2/ERF transcription factor OsRAV1 (RELATED TO ABI3 AND VP1) positively regulates vigor, salt tolerance. Additionally, was differently expressed embryo endosperm, with localized nucleus. Transcriptomic analysis revealed modulates through plant hormone signal transduction phenylpropanoid biosynthesis during germination. Haplotype showed rice varieties carrying Hap3 displayed enhanced tolerance These findings suggest potential target breeding high suitable cultivation.

The main objective of this review to explore the physiological molecular mechanisms governing rice seed under aerobic (water stress) anaerobic (hypoxic) conditions in direct-seeded (DSR) systems. Strong seedling vigor is imperative to achieve stable establishment and enhance the competitiveness against weeds in rice direct seeding. Shoot length (SL) one of important traits associated with rice, but few genes for SL have been cloned so far. In previous study, we identified two tightly linked stably expressed QTLs SL, qSL-1f qSL-1d by genome-wide association causal gene (LOC_Os01g68500) underlying qSL-1f. present identify LOC_Os01g66100 (i.e. semidwarf SD1), a well-known controlling plant height (PH) at adult-plant stage, as through gene-based haplotype analysis knockout transgenic verification. By measuring phenotypes (SL PH) various haplotypes their lines, found SD1 LOC_ Os01g68500 controlled both PH, worked same direction, which provided directly genetic evidence positive correlation between PH combined diverse natural population. Moreover, experiments suggested that had greater effect on compared Os01g68500, no significant difference SL. Further investigation pyramiding effects LOC_Os01g68500 based combinations may play dominant role when coexist. this stage validated. total, genes, are our studies, suitable beneficial desired different breeding objectives. These results provide new clue develop varieties seeding resources molecular strong vigor.

Strong early growth vigor is an essential target in both direct seeded rice breeding and high-yielding for varieties with relatively short duration the double-cropping region. Shoot dry weight (SDW) one of important traits associated vigor, breeders have been working to improve this trait. Finding stable QTLs or functional genes SDW crucial improving by implementing molecular rice.
