Embryonic genome activation is divided into a minor and major wave of transition to endogenous transcription. In bovines, begins early in the 2-cell stage completed by 8-cell when becomes dominant. While genes known regulate development have been studied extensively, involved more central cellular functions not examined. Taking advantage CRISPR Cas9 system, present study investigated effect knocking out Golgi retrograde protein transporter COPA on bovine development. After electroporation presumptive zygotes with ribonucleoproteins targeting exon 6, sequences 2 (11 %) 4-cell (16 embryos showed knockouts whereas blastocysts did not, demonstrating that necessary for beyond. Using repair template containing silent mutations along target site, loss wildtype was observed 5 blastocysts, successful knock-in at least one allele.
