The blood-brain barrier (BBB) is a crucial structure that maintains brain homeostasis by regulating the entry of molecules and cells from bloodstream into central nervous system (CNS). Neurodegenerative diseases such as Alzheimer's Parkinson's disease, well ischemic stroke, compromise integrity BBB. This leads to increased permeability infiltration harmful substances, thereby accelerating neurodegeneration. In this review, we explore mechanisms underlying BBB disruption, including oxidative stress, neuroinflammation, vascular dysfunction, loss tight junction integrity, in patients with neurodegenerative diseases. We discuss how breakdown contributes neurotoxicity, abnormal accumulation pathological proteins, all which exacerbate neuronal damage facilitate disease progression. Furthermore, potential therapeutic strategies aimed at preserving or restoring function, anti-inflammatory treatments, antioxidant therapies, approaches enhance integrity. Given role neurodegeneration, maintaining its represents promising approach slow prevent progression

Rumex vesicarius L. Polygonaceae is a wildly grown plant in Egypt, North Africa, and Asia with wide traditional uses. Several studies reported its biological activities richness phytochemicals. This research addresses comprehensive metabolic profiling of the flowers, leaves, stems, roots via RP-HPLC-QTOF-MS MS/MS chemometrics. A total 60 metabolites were observed grouped into phenolic acids, flavonoids, phenols, terpenes, amino fatty organic sugars. Principal component analysis hierarchal cluster showed segregation different parts. Moreover, antioxidant capacity was determined several methods agreed previous results. Additionally, an silico approach molecular docking predominant bioactive employed against two targets, NADPH oxidase human peroxiredoxin 5 enzyme (PDB ID: 2CDU 1HD2) receptors, alongside ADME predictions. The modelling revealed that most approached molecules specifically binding tested enzymes, achieving high affinities. results confirmed R. stems are rich sources components. To our knowledge, this first giving prospect relevance development new naturally based antioxidants.

Currently, nutraceuticals and functional food/cosmeceutical sectors are seeking natural molecules to develop various types of phytopharmaceutical products. Flavonoids have been reported in antioxidant many medical/pharmacological activities. Monochoria angustifolia or Siam violet pearl medicinal plant is the newest species genus C. Presl, which long consumed as food herbal medicines. Though previous work showed that apigenin-7-O-glucoside most abundant phytochemical found this plant, report on anti-aging activity still lacking needs be filled in. The objective explore capacities using both silico vitro assessments. In addition, pharmacokinetic properties were predicted. Interestingly, results from analysis a similar trend potential agent against three enzymes. properties, such adsorption, distribution, metabolism, excretion toxicity (ADMET), compound also provided work. current study first Thai plant. However, safety efficacy future developed products clinical should determined future.

ABSTRACT Parkinson's disease (PD) is the most prevalent neurodegenerative disease. Previously, it was believed that aberrant iron metabolism, leading to ferroptosis due glutathione (GSH) depletion, excessive Ca 2+ influx, mitochondrial (mROS), and cytosolic (cROS) free reactive oxygen species in brain, a contributing factor PD. ADP‐ribose (ADPR), mROS, cROS activate TRPM2 cation channel. It yet unclear how contributes development of neuronal damage induced by rise Our aim this study examine function protective effect GSH dopaminergic human SH‐SY5Y cells had been exposed 1‐methyl 4‐phenylpyridinium (MPP) produce parkinsonism. The were divided into six groups: control, MPP, MPP + erastin, erastin ferrostatin‐1, (GSH), blocker (ACA). In groups, concentrations , ADPR‐induced current density, membrane dysfunction, cROS, lipid peroxidation, Zn Fe increased, although peroxidase, GSH, cell viability, number decreased. changes higher group than only. However, their modulated ACA groups. conclusion, increase death parkinsonism (MPP)‐induced attributed activation. By regulating oxidant/antioxidant balance, regulates channel activity lowers ferroptosis.

Objectives Dietary flavonoids, known for their antioxidant and anti-inflammatory properties, may play a role in frailty prevention, but comprehensive population-based studies are lacking. This study aimed to examine the associations between dietary flavonoid intakes prevalence of nationally representative sample U.S. adults, identify predominant subclasses contributing these associations. Methods Cross-sectional data from 12,152 adults aged ≥20 years participating National Health Nutrition Examination Survey (NHANES) 2007–2010 2017–2018 were analyzed. intake was assessed using two 24-h recalls. Frailty defined 49-item index. Weighted multivariate logistic regression models restricted cubic spline analyses employed investigate relationships prevalence. quantile sum (WQS) g-computation (qgcomp) used assess mixed effects subclasses. Results Higher total flavonoids (OR:0.79, 95% CI:0.65–0.95), anthocyanidins (OR:0.71, CI:0.58–0.88), flavanones (OR:0.74, CI:0.59–0.92), flavones (OR:0.76, CI:0.59–0.97), flavonols (OR:0.67, CI:0.56–0.81) significantly associated with lower after adjusting confounders. Non-linear inverse observed flavonols. The WQS model revealed that mixture inversely odds (OR: 0.58, CI: 0.48–0.71, p < 0.001), flavones, flavonols, as top contributors. qgcomp confirmed findings highlighted potential opposing among Conclusion analysis provides evidence higher intakes, particularly anthocyanidins, adults. These suggest flavonoid-rich diets be promising strategy warranting further investigation through prospective cohort randomized controlled trials.
