Abstract Objective. To better understand the role of thyroid hormones in regulating growth breast epithelial cells and estrogen-like effects these hormones, present study was conducted to investigate relationship between cancer autoimmune disorders southeast Iran women. Methods. In this case-control study, case group, all newly diagnosed pa­tients referred oncology clinics Zahedan city years 2021‒2022 were studied. The num­ber participants each group represented by 40 patients. control women without cancer. sampling method simple or available using nonprobability method. presence absence dysfunction checked thyroid-stimulating hormone (TSH), free thyroxin (FT4), triiodothyronine (FT3), anti-thyroid peroxidase (TPO), anti-thyroglobulin (Tg) tests. Results. mean age 47±11 years. risk subclinical hyperthyroid­ism higher subjects with about 8 times than healthy ones (OR=8.27). According value OR=1, developing disease same individuals as individuals. odds ratio hypothy­roidism could not be calculated because zero. Conclusion. results show that there are abnormal characteristics thy­roid gland patients compared group. incidence hyperthyroidism significantly increased However, no difference two groups.

The incidence of thyroid papillary carcinoma (PTC) is increasing annually, causing both physical and psychological pressure on patients. Therefore, early recognition specific interventions for PTC are crucial. objective this study to explore novel diagnostic marker precise intervention targets PTC. Based a weighted gene co-expression network analysis (WGCNA), relevant datasets from the Gene Expression Omnibus (GEO) Cancer Genome Atlas (TCGA) databases were collected. Enrichment was performed differentially expressed genes (DEGs) using Ontology (GO), Disease (DO), Kyoto Encyclopedia Genes Genomes (KEGG), Set Analysis (GSEA). Subsequently, three machine learning algorithms Least Absolute Shrinkage Selection Operator (LASSO), Support Vector Machine Recursive Feature Elimination (SVM-RFE), Random Forest (RF) used identify core genes. Finally, receiver operating characteristic (ROC) curves analyze clinical value We found, in total, 11,194 DEGs derived TCGA GEO datasets, that primarily enriched extracellular matrix (ECM) inflammation related pathways, such as an ECM receptor interaction, cell adhesion molecules (CAMs), Tumor necrosis factor (TNF) signaling, nucleotide-binding oligomerization domain (NOD) like signaling pathways. Further genes, identified by protein-protein interaction network, discovered intersecting GJC1, KLHL4, NOL4. Of which, GJC1 has good ability, which verified (area under ROC curve (AUC) = .982) (AUC .840). highly it considered potential biomarker expected become new target therapy. However, still needs be supported more data.

Abstract The management of thyroid nodules is guided by the cytological classification provided Bethesda System for Reporting Thyroid Cytology. Notably, biology tumors in pediatric patients differs from that adults, and there limited research focused on cases. This study aimed to assess effectiveness system treated at largest tertiary center Poland between 2015 2023. A retrospective analysis was conducted 566 with nodules, whom 555 underwent fine-needle aspiration biopsy (FNAB). total 217 surgery. Of these, 206 had previously undergone FNAB evaluation our center, while 11 surgery due a RET mutation or need an extended procedure. initial results showed distribution across categories as follows: 7.6% category I, 54.6% II, 20.9% III, 4.1% IV, V, 5.6% VI. Among who surgery, I through VI 2.9%, 25.2%, 29.1%, 8.3%, 19.4%, 15%, respectively. risk malignancy (ROM) estimated 33.3%, 11.5%, 22.2%, 4.8%, 84.4%, 96.8% VI, In autoimmune thyroiditis (AIT), ROM higher than non-AIT it lower sensitivity detecting non-benign neoplasms III approximately 86% both AIT patients. However, papillary carcinoma, V but decreased 61.5% These findings emphasize importance considering surgical intervention III-VI cytology, particularly those AIT.

Abstract Background Pyroptosis, an inflammatory form of programmed cell death, has been implicated in the pathogenesis and progression several cancers. However, significance pyroptosis-related genes (PRGs) papillary thyroid cancer (PTC) remains unclear. Methods Transcriptome clinical data PTC patients were obtained from The Cancer Genome Atlas. expression patterns PRGs identified by consensus clustering. A prognostic model for predicting cancer-free interval (TCFi) employed five machine learning methods. Enrichment immune-related analyses performed to elucidate role pyroptosis. responses radioactive iodine (RAI), immune checkpoint inhibitors (ICIs), molecular targeted therapy (MTT), chemotherapy (CTx) predicted based on pyroptosis-derived features. Additionally, was validated via six external datasets, 16 lines, 20 pairs samples. Results classified into three PyroClusters, C1 exhibited BRFA-like tumors with highest invasiveness worst prognosis, C2 presented RAS-like tumors, C3 characterized gene fusion. Nine (CXCL8, GJA1, H2BC8, IFI27, PRDM1, PYCARD, SEZ6L2, SIGLEC15, TRAF6) filtered out construct a PyroScore model. derived nomogram demonstrated superior predictive performance than four staging systems. strong correlation between pyroptosis tumor microenvironment (TIME) remodeling observed mechanistic analyses. Patients high “hot” immunophenotypes had poorer prognosis but could benefit more ICIs CTx (such as paclitaxel). low sensitive RAI MTT pazopanib sorafenib). Abstract To investigate the expression, clinical significance, mechanism of action, and tumor microenvironment (TME) eight subunits chaperonin-containing TCP1 complex (CCT/TRiC) in thyroid cancer (TC). Methods :The Cancer Genome Atlas (TCGA) high-throughput functional genome (GEO) databases were used to analyze differential expression each CCT subunit TC normal tissues, TCGA Gene Expression Profiling Interaction Analysis (GEPIA) effects levels on patient prognosis construction survival risk models. Enrichment (GSEA) was subunit's biological functions, microenvironment, drug sensitivity, immune checkpoints analyzed using Tumor Immune Infiltration Database (TIMER2.0). Further analysis performed correlation between cell infiltration TCs targets. Results :There a significant difference found tissues ( P < 0.05). The affect are closely related TME, infiltration, checkpoints, sensitivity. Conclusions :CCT complexes potential molecular biomarkers for cancer, which may promote development by affecting thus influencing prognosis. They also have serve as targets diagnosis immunotherapy refractory cancer.
