Identification of GJC1 as a novel diagnostic marker for papillary thyroid carcinoma using weighted gene co-expression network analysis and machine learning algorithm

The
incidence
of
thyroid
papillary
carcinoma
(PTC)
is
increasing
annually,
causing
both
physical
and
psychological
pressure
on
patients.
Therefore,
early
recognition
specific
interventions
for
PTC
are
crucial.
objective
this
study
to
explore
novel
diagnostic
marker
precise
intervention
targets
PTC.
Based
a
weighted
gene
co-expression
network
analysis
(WGCNA),
relevant
datasets
from
the
Gene
Expression
Omnibus
(GEO)
Cancer
Genome
Atlas
(TCGA)
databases
were
collected.
Enrichment
was
performed
differentially
expressed
genes
(DEGs)
using
Ontology
(GO),
Disease
(DO),
Kyoto
Encyclopedia
Genes
Genomes
(KEGG),
Set
Analysis
(GSEA).
Subsequently,
three
machine
learning
algorithms
Least
Absolute
Shrinkage
Selection
Operator
(LASSO),
Support
Vector
Machine
Recursive
Feature
Elimination
(SVM-RFE),
Random
Forest
(RF)
used
identify
core
genes.
Finally,
receiver
operating
characteristic
(ROC)
curves
analyze
clinical

total,
11,194
DEGs
derived
TCGA
GEO
datasets,
that
primarily
enriched
extracellular
matrix
(ECM)
inflammation
related
pathways,
such
as
an
ECM
receptor
interaction,
cell
adhesion
molecules
(CAMs),
Tumor
necrosis
factor
(TNF)
signaling,
nucleotide-binding
oligomerization
domain
(NOD)
like
signaling
pathways.
Further
genes,
identified
by
protein-protein
interaction
network,
discovered
intersecting
GJC1,
KLHL4,
NOL4.
Of
which,
GJC1
has
good
ability,
which
verified
(area
under
ROC
curve
(AUC)
=

Abstract
The
management
of
thyroid
nodules
is
guided
by
the
cytological
classification
provided
Bethesda
System
for
Reporting
Thyroid
Cytology.
Notably,
biology
tumors
in
pediatric
patients
differs
from
that
adults,
and
there
limited
research
focused
on
cases.
This
study
aimed
to
assess
effectiveness
system

at
largest
tertiary
center

between
2015
2023.

Background
Pyroptosis,

inflammatory

Abstract

Conclusions
