Introduction. Fungi can synthesize a diverse range of melanins with appropriate physicochemical and biological characteristics for numerous applications in health, environmental protection, energy, industry. Gaining deeper insights into the chemical structures, biosynthetic pathways, regulatory mechanisms fungal melanin would establish basis metabolic engineering approaches, aimed at enhancing production efficiency creating custom-designed desirable material properties. Dihydroxynapthalene-(DHN) and L-dihydroxyphenylalanine (L-DOPA) are two types of dominant melanin in fungi. Fungal melanins with versatile functions frequently associated pathogenicity stress tolerance. In rice blast fungus, Magnaporthe oryzae, DHN is essential to maintain the integrity infectious structure, appressoria; but role tyrosinase-derived L-DOPA still unknown. Here, we have genetically biologically characterized a tyrosinase gene (MoTyr) M. oryzae. MoTyr encodes protein 719 amino acids that contains typical CuA CuB domains tyrosinase. The deletion mutant (ΔMoTyr) was obtained by using homologous recombination approach. Phenotypic analysis showed conidiophore stalks conidia formation significantly reduced ΔMoTyr. Under different concentrations glycerol PEG, more appressoria collapsed strains than wild type, suggesting appressorium wall. Melanin measurement confirmed loss resulted significant decrease synthesis. Accordingly, stunted germination under conditions. Importantly, affected both infection pathogenesis stages. These results suggest MoTyr, like pigment synthase, plays key formation, integrity, revealing potential drug target for disease control.

Melanin is one of the most abundant natural biomolecules on Earth. These macromolecular biopolymers display several unique physical and chemical properties have garnered interest as biomaterials for various commercial industrial applications. To this end, extensive research has gone into refining methods synthesis extraction melanin from recombinant sources. In study, we developed refined a procedure using microbial system biosynthesis tyrosinase enzyme Tyr1 tyrosine substrate. Using emergent chassis organisms Vibrio natriegens , achieved maximal yields 7.57 g/L, highest reported volumetric productivities 473 mg L −1 h with 100% conversion rates in an optimized, minimally defined medium. Morchella sextelata , a highly sought-after edible mushroom worldwide, is evaluated based on its cap color as an essential commercial property indicator.
