sp. PM06, previously isolated from sugarcane waste pressmud, could produce dual enzymes α-amylase and cellulase. The isolate's crude were purified homogeneously using ammonium sulfate precipitation followed by High Quaternary amine anion exchange chromatography. Purified revealed the molecular weights of cellulase as 55 52 kDa, with a purification fold 15.4 11.5, respectively. specific activity 740.7 555.6 U/mg, It demonstrated wide range pH 5.0 to 8.5, an optimum 5.5 6.4 for temperature was 50 °C 60 kinetic parameters 741.5 ± 3.75 µmol/min/mg

The interest in microbial pectinases is increasing due to their use the biotechnological and food industries response global demand. In this study, among 22 halophilic bacterial strains screened, 20 showed pectinase activity. highest activity was measured for Virgibacillus salarius strain 434. As production meets many parameter optimization constraints, optimal conditions maximizing yield of synthesized using investigated were followed. Enzyme purification accomplished at up 11.5-fold via ultrafiltration gel filtration chromatography, until enzyme’s specific had increased 10-fold (104.3 U/mg). molecular weight determined be 68 kDa. Km Vmax values 0.38 mg/mL 120 U/mg, respectively, determined. purified from 434 good thermal stability. A half-life min 70 °C high catalytic under haloalkaline (pH 9 g L−1 NaCl) observed. Its storage stability −20 exceeded 2 months. This demonstrates its potential application where these extremes are present, as harsh industrial processes require novel with unique properties instead hazardous chemicals.
