Abstract Limbs are a defining characteristic of tetrapods, yet numerous taxa, primarily among amphibians and reptiles, have independently lost limbs as an adaptation to new ecological niches. To elucidate the genetic factors contributing this convergent limb loss, we present 12 Gb chromosome-level assembly Banna caecilian (Ichthyophis bannanicus), limbless amphibian. Our comparative analysis, which includes reconstruction amphibian karyotype evolution, reveals constrained gene length evolution in subset developmental genes across 3 large genomes. Investigation development uncovered loss Grem1 caecilians Tulp3 snakes. Interestingly, snakes share significantly larger number degenerated conserved noncoding elements than lizards, shorter evolutionary history loss. These overlap with active genomic regions during mouse limbed species, suggesting their essential role patterning tetrapod common ancestor. While most emerged jawed vertebrate ancestor, coinciding origin paired appendage, more recent also contribute development, demonstrated through functional experiments. study provides novel insights into regulatory associated offering perspective on basis morphological specialization.

Reindeer have long been served as vital subsistence resources for inhabitants of Arctic and subarctic regions owing to their domestication. However, the evolutionary relationships divergence times among different reindeer populations, genetic traits that distinguish domesticated reindeer, factors contribute relative docility compared with other Cervidae specie, remain unclear. In this study, we sequenced genomes 32 individuals from wild domestic populations inhabit regions. We found experienced 2 or more independent domestication events characterized by weak artificial selection pressure limited significant differences in genomic parameters between populations. Alterations conserved noncoding elements genomes, particularly those associated nervous system development, may contributed rendering less responsive. Together, our results suggest inherent species-specific traits, rather than intense selection, played a role relatively docile behavior offer valuable insights into process these animals.

The gold barb (Barbodes semifasciolatus), a member of the Cyprinidae family, exhibits remarkable adaptability to highly acidic environments, making it an ideal model for studying extreme environmental adaptation. However, its genome has not been previously characterized. To address this, we assembled high-quality chromosome-scale B. semifasciolatus using High-Fidelity (HiFi) sequencing and Hi-C technology. resulting haplotype-resolved assemblies, spanning 776 Mb 779 across 25 chromosomes, achieved coverages 99.5% 99.7%, respectively, included four gap-free chromosomes. Genome quality assessment BUSCO indicated high completeness score 98.2% haplotype1 98.3% haplotype2, further validated by strong synteny with zebrafish (Danio rerio), confirming assembly's integrity continuity. Through integration full-length transcriptome data, RNA sequencing, homology-based annotation, identified 26,057 protein-coding genes 2,087 pseudogenes in haplotype 2, 25,622 2,101 1. This high-resolution assembly is crucial resource advancing research Cyprinidae, particularly understanding adaptive evolution environments.
