Arkadia/RNF111 large RING ligase modifies some substrates degradative chains, while other are modified non-degradative Here, using Arkadia related ligase, Ark2C, we characterize RING-E2 complexes promote assembly distinct linkages. Our structural studies highlight conserved interface, our binding activity assays reveal several E2 enzymes functionally interact Ark2C Arkadia. Using Arkadia, ubiquitylation differences ability be modified, Ubc13 Ube2K requiring addition 'priming' molecule before subsequent can occur.
