Parkinson's
disease
(PD)
is
a
neurodegenerative
characterized
by
the
gradual
loss
of
dopaminergic
neurons
in
substantia
nigra
pars
compacta
(SNpc),
abnormal
accumulation
α-synuclein
(α-syn),
and
activation
microglia
leading
to
neuroinflammation.
Disturbances
circadian
rhythm
play
significant
role
PD,
with
most
non-motor
symptoms
associated
disruptions
rhythm.
These
disturbances
can
be
observed
years
before
motor
appear
are
marked
emergence
related
such
as
rapid
eye
movement
sleep
behavior
disorder
(RBD),
restless
leg
syndrome
(RLS),
excessive
daytime
sleepiness
(EDS),
depression
anxiety,
changes
blood
pressure,
gastrointestinal
dysfunction,
urinary
problems.
Circadian
disruption
precedes
onset
contributes
progression
PD.
In
brief,
this
article
outlines
triggering
PD
at
cellular
molecular
levels,
well
its
clinical
manifestations.

Alpha-synuclein
(α-syn)
is
a
140-amino-acid,
intrinsically
disordered,
soluble
protein
that
abundantly
present
in
the
brain.
It
plays
crucial
role
maintaining
cellular
structures
and
organelle
functions,
particularly
supporting
synaptic
plasticity
regulating
neurotransmitter
turnover.
However,
for
reasons
not
yet
fully
understood,
α-syn
can
lose
its
physiological
begin
to
aggregate.
This
altered
disrupts
dopaminergic
transmission
causes
both
presynaptic
postsynaptic
dysfunction,
ultimately
leading
cell
death.
A
group
of
neurodegenerative
diseases
known
as
α-synucleinopathies
characterized
by
intracellular
accumulation
deposits
specific
neuronal
glial
cells
within
certain
brain
regions.
