The prevalence of lipid metabolism disorders, including obesity, increases with age in cats and humans. Obesity is a condition characterized by systemic low-grade inflammation oxidative stress caused excessive visceral fat accumulation. Resveratrol (RSV), natural plant polyphenol, modulates the expression anti-inflammatory factors. This study aimed to investigate effects resveratrol supplementation on both healthy obese assess its potential as dietary supplement for improving disorders this population. Plasma metabolite hormone concentrations, enzyme activities were measured healthy, obese, overweight supplemented RSV 4 weeks. RVS was at 1 mg/kg body weight/day (low dose) 5 mg/kg/day (high capsules Body weight, score, BUN, insulin concentrations did not change or triglyceride, free fatty acids, serum amyloid A (SAA) lactate dehydrogenase (LDH) decreased, adiponectin increased markedly after supplementation. Decreased plasma SAA LDH seem be induced improvement liver function effect RSV. Moreover, may useful treating cats.

Liver disease is a significant global health issue, responsible for millions of deaths annually. Aging, characterized by the gradual decline in cellular and physiological functions, impairs tissue regeneration, increases susceptibility to liver diseases, leads health. Silent information regulator 1 (SIRT1), NAD⁺-dependent deacetylase, has emerged as pivotal factor modulating age-related changes liver. SIRT1 preserves function regulating essential aging-related pathways, including telomere maintenance, epigenetic modifications, senescence, intercellular communication, inflammation, mitochondrial function. Notably, levels naturally with age, contributing progression increased vulnerability injury. This review summarizes regulatory role aging its impact on diseases such fibrosis, alcoholic associated (ALD), metabolic dysfunction-associated steatotic (MASLD), steatohepatitis (MASH), hepatocellular carcinoma (HCC). We also discuss emerging therapeutic approaches, activators, gene therapy, nutritional interventions, which are evaluated their potential restore mitigate progression. Finally, we highlight future research directions optimize SIRT1-targeted therapies clinical applications conditions.

ABSTRACT Resveratrol alleviates liver fibrosis in mice by upregulating IL‐10 to reprogram the macrophage phenotype; however, mechanism remains be elucidated. Building on our previous work, this study, we aimed determine role of TLR2/MyD88/ERK and NF‐κB/NLRP3 inflammasome pathways mediating effects resveratrol polarization. We investigated expression cytokines these a mouse model resveratrol‐treated macrophages. The results showed that TLR2, MyD88, ERK, NF‐κB1 tissues was increased fourth week after treatment with but decreased fifth week. Similar were obtained for pathway. also both macrophages elevated 24 h reduced 36 resveratrol. Immunofluorescence nucleocytoplasmic separation assays inhibited translocation NF‐κB from cytoplasm nucleus macrophages, associated inhibition Silencing ERK. In conclusion, are involved effect polarization subsequent modulation fibrosis. acts as common cytokine coordinates crosstalk between two pathways. Given the growing challenges posed by an ageing population, particularly in Western countries, we aimed to investigate potential geroprotective effects of resveratrol and melatonin rats. The animals were treated with these two compounds starting at 3 months age continuing until 1 year or 2 years age. Using a multibiomarker approach, assessed DNA damage, telomere length, oxidative status their urine, liver, kidneys. Despite employing this experimental our results did not provide conclusive evidence across evaluated organs. However, observed sex-dependent differences response treatment. high potency compounds, further research is warranted explore incorporation into daily routines as strategy mitigate ageing-related effects.

Long-term exposure to arsenic, a prevalent environmental contaminant, has been implicated in the pathogenesis of various hepatic conditions. Hepatic stellate cells (HSCs) are central development liver fibrosis. Recently, involvement interleukin-17 (IL-17) and NOD-like receptor protein 3 (NLRP3) inflammasome pathologies attracted significant research interest. Hepatocyte pyroptosis, form programmed cell death, is critical factor occurrence inflammation. The objective this study was investigate specific roles IL-17 NLRP3 arsenic-induced activation HSCs through hepatocyte pyroptosis. We pretreated MIHA with MCC950 (1 5 μM) secukinumab (10 100 nM) for 4 h, then NaAsO2 (25 24 h at 37 °C under 5% CO2. After incubation, cell-culture supernatant collected mixed serum-free high-glucose DMEM medium 1:1 ratio prepare conditioned medium, which subsequently used culture LX-2 cells. results showed that induced hepatocellular led release inflammatory cytokines IL-18 IL-1β subsequent HSCs. Treatment inhibitors significantly reduced secretion Extracellular matrix (ECM) components attenuated HSC activation. These demonstrate blocking signaling pathways reduces attenuates fibrogenesis. provide novel molecular targets prevention treatment arsenic-related
