In vitro cardiomyocyte mechano-sensing platform is crucial for evaluating the mechanical performance of cardiac tissues and will be an indispensable tool application in drug discovery disease mechanism study. Magnetic sensing offers significant advantages real-time, situ wireless monitoring resistance to ion interference. However, due mismatch between stiffness traditional rigid magnetic material myocardial tissue, sensitivity insufficient it difficult achieve cell structure induction three-dimensional cultivation. Herein, a that integrates neodymium–iron–boron/polydimethylsiloxane (NdFeB/PDMS) flexible microbeam with suspended ordered polycaprolactone (PCL) nanofiber membranes was developed, providing anisotropic culture environment growth simultaneously realizing contractility monitoring. The as-prepared sensor presented ultrahigh 442.2 μV/μm deflection resolution 2 μm. By continuously status stimulation feedback, we verified capability capture dynamic changes contractility. Globally, a number of diseases impact us and while treatment options exist, it is often found that similar treatments have variable effects on different patients with the same disease. Particularly in case conditions are closely associated genetics (like cancer), intensity results vary between patients. Even for like arthritis not uncommon only fraction to achieve remission therapeutic approach. With millions suffering from cancer arthritis, precision medicine (PM) has been at forefront biomedical pharmaceutical research since 2015. PM focusses understanding genetic environmental factors affecting several platforms. One platforms use three-dimensional (3D) vitro models, especially those derived patient themselves. These organ-on-chip (OOC), organoid spheroid 3D biomaterial scaffolds others, advantages over traditional two-dimensional (2D) cell culture approaches. In this opinion paper, author briefly discusses used PM. Then, models 2D vivo considered an overview their applications provided. A thorough characterization of induced pluripotent stem cells (iPSCs) used with in vitro models or therapeutics is essential. Even iPSCs derived from a single donor can exhibit variability within and between cell lines, which lead to heterogeneity results hinder the promising future replacement therapies. In this study, seeding density human rhesus monkey was tested maximize line-specific yield generated cardiomyocytes. We found that, despite using same iPSC generation differentiation protocols, for best efficiency could differ by factor four lines here. addition, showed differences range densities that they tolerate without severe loss efficiency. Overall, our data show critical parameter inefficiency primate cardiomyocytes episomal approach still considerable heterogeneity.
