Prenatal particulate matter exposure imprints an epigenetic scar on FCER1G in fetal macrophages, reemerging in M2 macrophages to drive atopic dermatitis development

Prenatal environmental exposures may influence the development of atopic dermatitis (AD), yet underlying mechanisms remain poorly understood. In this study, we conducted an epidemiological analysis 986 children, revealing that first-trimester PM_2.5 exposure significantly increases AD risk at age three. Integrated epigenomic and transcriptomic profiling 48 placentae identified PM_2.5-associated hypomethylation driving FCER1G overexpression, predominantly in fetal macrophages. This "epigenetic scar" persists across developmental stages, as demonstrated through single-cell RNA sequencing 404,169 cells from placenta, fetal, adult skin samples. skin, was progressively overexpressed M2 macrophages, particularly within AD-specific cell states. Network vitro PM experiments a mechanistic axis involving FCER1G, CD68, CYBB, MRC1, MS4A4A, promoting NADPH oxidase–driven reactive oxygen species signaling. Our findings establish link between prenatal pathogenesis persistent macrophage dysregulation, highlighting placental epigenetic scars critical mediators immune target for intervention.

Early life exposures of childhood asthma and allergies—an epidemiologic perspective

Children around the world are continuing to develop and suffer from chronic lung diseases such as asthma. Childhood asthma commonly presents with recurrent episodes of cough, shortness breath, wheezing, all which can lead missed school days hospitalization admissions. The role environmental pollutants aeroallergens has been increasingly recognized in relation etiology. We showcase impacts air pollution pollen exposures early life on childhood allergies through an epidemiologic perspective. also examine effects indoor microbial endotoxin glucan allergic schoolchildren many spend most their time a household or classroom setting. Findings this work assist identification key factors critical periods improve clinicians’ diagnoses during childhood.

Risk factors of allergic rhinitis and its prevention strategies

Allergic rhinitis (AR) is a global disease with high prevalence. It reduces the patient's quality of life seriously. The health care and management AR was also heavy social burden. Specific immunotherapy (SIT) only curative treatment for that may alter natural course this disease. However, acceptance compliance SIT in patients are still not many effectively controlled. Disease prevention based on known risk factors much more cost-effective compared to post-diagnosis treatment. There have been some reports up now, but information fragmented. This review systemically clarified including hereditary family history, maternal situation & mode delivery feeding, personal characteristics, nutrition food intake, behavior habits, acquired environmental chemical exposure, diseases status. preventive strategies were proposed briefly. hopeful improve people's awareness put forward prevention.
