Psoriasis is one of the most common chronic inflammatory skin diseases. Many studies suggest that dendritic cells (DCs) and T cell-mediated interleukin (IL)-23/IL-17 axis play a central role in signaling pathway pathogenesis psoriasis. Chlorella, also known as Parachlorella beijerinckii (PB), unicellular green alga has long been used health food. It contains carotenoids have antioxidant anti-inflammatory effects. In this study, we investigated whether PB-derived (PBCs) ameliorated processes an imiquimod (IMQ)-induced psoriasis-like mouse model bone marrow-derived (BMDCs). We found PBCs attenuated erythema, thickness, scaling, neutrophil infiltration tissue IMQ-induced mice. Moreover, suppressed psoriasis-related pro-inflammatory cytokine expression, DC activation, IL-17A production by γδ BMDCs, expression levels cytokines, including IL-23; IL-1β; IL-6; CD40/CD86, marker activation. Additionally, inhibited nuclear factor kappa B, p38, c-Jun NH2-terminal kinase pathways mitochondrial reactive oxygen species (mitoROS)-triggered inflammasome activation pathway. activated extracellular regulated protein kinase/NF-E2-related factor-2 (ERK/Nrf2) BMDCs. harmful effects gene mitoROS via ERK/Nrf2 conclusion, may be beneficial management psoriatic inflammation.

Vascular cell adhesion molecule-1 (VCAM-1) and E-selectin are involved in different inflammatory diseases may be potential cardiovascular risk biomarkers psoriasis. They play an important role regulating the recruitment to endothelial cells during inflammation, affecting various conditions like vasculitis, atherosclerosis, diseases. Positive outcomes have been observed when using Tumor Necrosis Factor Alpha (TNF-α) inhibitors biological therapies that target selectins control functioning of reduce inflammation psoriasis related conditions. Moreover, effects systemic treatments ultraviolet B (UVB) phototherapy on VCAM-1 levels patients highlights impact severity activation cells. In addition, factors such as age, sex, metabolic syndrome, hyperglycemia, migraines, tobacco smoking found affect E-selectin. This sheds light understanding complex relationship between development Studies show indicators treatment effectiveness progression disease. summary, this review importance for assessing disease individuals with The shared mechanisms along effect markers, provide significant insights further research approaches manage future.

Chronic rhinosinusitis (CRS) is a prevalent inflammatory airway disease affecting over 10% of the global population, leading to considerable socio-economic impacts, especially in developing countries. The pathogenesis CRS multifactorial, involving potential contributions from both genetic and environmental factors. While influence allergic autoimmune diseases on has been observed, causal relationships between these remain unclear. We extracted data large-scale genome-wide association studies (GWAS) utilized bidirectional two-sample Mendelian randomization (MR) analysis explore ten diseases, including asthma, rhinitis (AR), atopic dermatitis (AD), psoriasis, type 1 diabetes (T1D), hypothyroidism, celiac (CeD), multiple sclerosis (MS), rheumatoid arthritis (RA), systemic lupus erythematosus (SLE). Additionally, we conducted colocalization determine whether allergic/autoimmune showing statistical with are driven by same variants. MR identified that AR (OR = 1.30; 95% CI 1.21-1.40; P 3.26E-13), asthma 1.35; 1.25-1.45; 1.35E-14), AD 1.17; 1.06-1.30; 0.003) were significantly associated an increased risk CRS. Interestingly, psoriasis 0.05; 0.01-0.37; 0.004) appeared have protective effect against Associations for T1D hypothyroidism also suggestive as factors No significant associations reverse analysis, suggesting one-directional relationship. Colocalization indicated (PP.H4 0.99) shared variant (IL-33 rs3939286) In conclusion, our study confirmed (AR, AD, psoriasis) Notably, variant, rs3939286 IL-33 gene, CRS, targeting pathway may provide therapeutic strategy diseases.

Psoriasis is a chronic inflammatory disease with complex pathogenesis, influenced by various factors involving environment, genes, and immunity. The main symptoms of psoriasis include erythema, scales, itching, etc. At present, therapeutic drugs for are continually evolving towards enhancing treatment efficacy reducing side effects. Firstly, the pathogenesis characteristics were summarized. Then, types benefits topical therapy introduced, such as aspects avoiding systemic toxic effects, first pass effect, gastrointestinal reactions accelerating onset time improving its efficacy, compared to drugs. In case methotrexate, cyclosporin A, Janus kinase (JAK) inhibitors, phosphodiesterase-4 (PDE-4) this review had further discussion on improvement translation these molecules from in clinical practice. To augment limitation skin permeability, nanotechnology novel drug delivery system including nanomedicines, hydrogels, ionic liquids, microneedles elaborated management. Also, exploration targeting pathogenic genes through small interfering RNA (siRNA) using nanoparticles liquids (ILs) great significance long-term psoriasis. Taken together, development numerous platforms expected achieve enhanced penetration, precise efficient molecule interference (RNAi) therapeutics prospects.

ABSTRACT Fibroblasts (FBs) are crucial mesenchymal cells that preserve the skin's natural structure and physiological processes. They can build dense connective tissue by remodelling extracellular matrix (ECM) control immunological activity secreting cytokines. This indicates development of chronic inflammatory proliferative skin disorders is significantly influenced fibroblasts. In order to provide new ideas for clinical research treatment with a clearer perspective, this study thoroughly compiles involvement fibroblasts in various diseases such as psoriasis, hypertrophic scar (HS), keloid, atopic dermatitis (AD), oral lichen planus (OLP), eczema, rosacea.
