Microglia are resident immune cells in the central nervous system that rapidly activated to mediate neuroinflammation and apoptosis, thereby aggravating brain tissue damage after ischemic stroke (IS). Although scutellarin has a specific therapeutic effect on IS, potential target mechanism of its treatment not been fully elucidated. In this study, we explored treating IS using network pharmacology. Lipopolysaccharide (LPS) was used induce an vitro BV-2 microglial cell model, while middle cerebral artery occlusion (MCAO) vivo animal model. Our findings indicated promoted recovery blood flow MCAO rats at 3 days, significantly different from group. Western blotting immunofluorescence revealed resulted significant reduction protein expression levels incidence immunopositive for p-NF-

Background: Clinical and experimental evidence indicates that copper has the ability to promote progressive development of demyelinating diseases such as multiple sclerosis. Microglia-mediated neuroinflammation is believed play a crucial role in this process. Scutellarin, flavonoid compound, anti-inflammatory, antioxidative, neuroprotective effects. Aim: We investigated effect scutellarin on copper-induced inflammatory foam cell formation microglia. Methods: exposed BV2 murine microglial cells copper, then collected conditioned medium co-cultured it with MO3.13 human glial mimic myelin damage vitro. The Cell Counting kit-8 assay, quantitative (polymerase chain reaction) PCR, enzyme-linked immunosorbent Luxol fast blue staining, western blotting were used detect phenotype. To investigate whether exposure can cause neurotoxicity indirect cells, we determined inflammation through by oil red O staining detection malondialdehyde (MDA) content. Finally, treated its therapeutic Results: Exposure activated pro-inflammatory phenotype microglia, assessed measuring transcription M1/M2-related biomarkers. In addition, increased intake microglia promoted intracellular lipid accumulation oxidation, facilitating formation. Rescue experiments showed chelator ammonium tetrathiomolybdate (ATTM) oxidation inhibitor ferrostatin-1 (Fer-1) significantly inhibited inflammation, reduced MDA levels, decreased Moreover, phosphorylation p38 mitogen-activated protein kinase (MAPK) led shift towards M1 transformation, which effectively ATTM, Fer-1, MAPK SB203580. Lastly, after treatment scutellarin, exhibited phosphorylation, production neurotrophic factors, expression mediators, accumulation, induced polarization M2 Conclusions: Here, demonstrated induce myelinating key mechanism involving MAPK. Scutellarin partially reversed positive effects promoting polarization, deposition, mediating signaling pathway. Taken together, these results suggest may be promising drug for

Erigeron breviscapus exhibits anti-inflammatory properties, protects neuronal cells and enhances immune function. Modern traditional Chinese medicine fermentation techniques can increase the bioactive compound content in breviscapus. However, its potential therapeutic effects against porcine pseudorabies virus (PRV) remain unclear. A PRV infection model was established mouse trigeminal ganglion (TG) to determine optimal antiviral mode of action flavonoids from fermented (FEBF). Additionally, a PRV-infected rat developed evaluate vivo efficacy FEBF. FEBF demonstrated higher protective rate lower viral copy number compared unfermented E. (EBF). The effect most pronounced under toxicological inhibitory conditions, surpassing blocking effect. upregulated TLR4, MyD88, NF-κB p65 protein expression during pre-infection phase, followed by their downregulation after 12 h. regulated PRV-induced changes expression, restoring them near-normal levels 36 In assessments pathological injury, load, count, apoptosis indicated that provided superior neuroprotection both Minocycline (MINO), broad-spectrum neuroprotective drug, EBF. Mechanistic studies further revealed modulated microglial polarization inflammatory cytokines IL-6, TNF-α, IL-4, IL-10. These findings demonstrate significant vitro models. represents promising candidate for development anti-PRV therapeutics.

Ischemic stroke (IS), primarily caused by cerebrovascular obstruction, results in severe neurological deficits and has emerged as a leading cause of death disability worldwide. Recently, there been increasing exploration the neuroprotective properties inert gas argon. Argon exhibited impressive neuroprotection many vivo ex experiments without signs adverse effects, coupled with advantages being inexpensive easily available. However, efficient administration strategy underlying mechanisms argon IS are still unclear. This review summarizes current research on effects goal to provide effective guidance for application elucidate potential neuroprotection. Early appropriate at high concentration possible offers favorable IS. inhalation shown some long-term protection benefits. provides anti-oxidative stress, anti-inflammatory anti-apoptotic cytoprotective mainly around Toll-like receptor 2/4 (TLR2/4), mediated extracellular signal-regulated kinase 1/2 (ERK1/2), nuclear factor (erythroid-derived 2)-like 2 (Nrf2), kappa-B (NF-ĸB) B-cell leukemia/lymphoma (Bcl-2). Therefore, holds significant promise novel clinical agent ischemic after further researches identify optimal mechanism.

Abstract Previous studies have shown that scutellarin inhibits the excessive activation of microglia, reduces neuronal apoptosis, and exerts neuroprotective effects. However, whether regulates activated microglia-mediated apoptosis its mechanisms remains unclear. This study aimed to investigate can attenuate PC12 cell induced by microglia via JAK2/STAT3 signalling pathway. Microglia were cultured in oxygen–glucose deprivation (OGD) medium, which acted as a conditioning medium (CM) activate cells, expression signalling-related proteins. We observed cells CM was significantly increased, fluorescence intensity pro-apoptotic protein Bax apoptosis-related cleaved caspase-3 anti-apoptotic B-cell lymphoma-2 (Bcl-2) decreased. Phosphorylation levels pathway-related proteins JAK2 STAT3 After treatment with scutellarin, well The Bcl-2, phosphorylated JAK2, increased. AG490, specific inhibitor pathway, used. Our findings suggest AG490 attenuates effects scutellarin. revealed inhibited OGD-activated regulated

Abstract Scutellarin is an herbal agent which can exert anti‐neuroinflammatory effects in activated microglia. However, it remains uncertain if inhibit microglia‐mediated neuroinflammation by regulating miRNAs. This study sought to elucidate the upstream regulatory mechanisms endogenous microRNAs and its target gene microglia lipopolysaccharide (LPS)‐induced BV‐2 Results show that scutellarin suppressed expression of tumor necrosis factor‐α (TNF‐α), interleukin‐1β (IL‐1β), inducible nitric oxide synthase (iNOS) significantly LPS‐stimulated As with results miRNAs function classification vitro, levels mir‐7036a‐5p are upregulated LPS‐activated microglia, but downregulated scutellarin. Rescue experiments indicated a pro‐inflammatory factor agomir promoted phosphorylated tau proteins (p‐tau), protein kinase C gamma type (PRKCG), extracellular regulated kinases (ERK1/2), reversed antagomir vitro. It shown here involved inflammation LPS‐induced More important novel finding mitigated down‐regulating mir‐7036a‐5p/MAPT/PRKCG/ERK signaling pathway.
