Aluminum (Al) is a ubiquitous element with known toxicity for both humans and animals. Herein, we aimed to investigate the potential role of melatonin (MEL) in hepatotoxicity nephrotoxicity following aluminum chloride (AlCl3) treatment rats. Adult male rats were treated AlCl3 (34 mg/kg bwt) eight weeks. Exposure enhanced serum activities liver transaminases (alanine aminotransferase aspartate aminotransferase) increased level bilirubin, addition kidney function markers urea creatinine. intoxication boosted oxidative stress, as evidenced by increases levels lipid peroxidation (LPO) nitric oxide (NO) along simultaneous decreases glutathione (GSH), various antioxidant enzymes, Nrf2 mRNA expression. MEL (5 repressed LPO NO levels, whereas it augmented GSH content. The enzymes GPx, SOD, CAT, GR also restored concomitantly when was administered before AlCl3. suppressed apoptotic effect enhancing Bcl-2 protein expression decreasing proinflammatory cytokines. Histopathological findings tissues confirmed beneficial against toxicity. These indicate that prevents defense system.

Exposure to aluminum chloride (AlCl3) induces progressive multiregional neurodegeneration in animal models by promoting oxidative stress and neuroinflammation. The current study was designed assess the potential efficacy of natural antioxidants celastrol thymoquinone (TQ) for alleviating AlCl3-induced psychomotor abnormalities oxidative-inflammatory burden male albino rats. Four treatment groups were compared: (i) a vehicle control group, (ii) AlCL3 group receiving daily intraperitoneal (i.p.) injection AlCl3 (10 mg/kg) 6 weeks, (iii) plus TQ mg/kg, i.p.) cotreatment (iv) (1 group. Open-field, rotarod, forced swimming tests conducted locomotor activity, motor coordination, anxiety-like behavior, depressive-like behavior. Acetylcholine (ACh), dopamine, serotonin levels measured brain homogenates. Malondialdehyde (MDA), total antioxidant capacity (TAC), catalase activity as markers, while tumor necrosis factor-α (TNF-α) interlukin-6 (IL-6) expression inflammatory markers. Brain derived neurotrophic factor (BDNF) mRNA an index endogenous neuroprotective response. Daily reduced free ambulation, impaired promoted anxiety- depression-like behaviors, whole-brain ACh, concentrations, increased MDA accumulation, TAC, elevated TNF-α IL-6, suppressed BDNF expression.
