Pharmacogenomics is the study of genetic modifications in patients that influence their potential to metabolize medications, encouraging tolerability and desired effect. Genetic (like genome-wide association & candidate gene) methods may help choosing most suitable drug dosage, by impeding adverse reactions (ADRs). are usually documented as key steps personalized medicine. The significance medicine healthcare developing due its defined molecular basis. Personalized anticipates beneficial or serious side effects a medication using specific individual's profile. In recent year's pharmacogenomics approaches being widely used personalizing therapies for diseases like Cancer, Rheumatoid arthritis, Asthma, Type-1 diabetes many psychiatric disorders (depression, psychosis, schizophrenia etc.).This chapter provides an overview present importance identifies challenges need be conquered effective

Ovarian cancer (OC) is a global health problem that frequently presents at advanced stages, predisposed to recurrence, readily develops resistance platinum-based drugs, and has low survival rate. Predictive, preventive, personalized medicine (PPPM/3PM) offers an integrated solution with the use of genetic, proteomic, metabolic biomarkers identify high-risk individuals for early detection. Metabolic reprogramming one key strategies employed by tumor cells adapt microenvironment support unlimited proliferation. Pyruvate kinases M1 M2 (PKM1/2) are encoded PKM gene, pivotal enzyme in last step glycolytic pathway, which crossroads aerobic oxidation Warburg effect serve as potential regulator glucose metabolism influence cellular energy production reprogramming. Commonly, ratio PKM1-to-PKM2 changed tumors compared normal controls, PKM2 highly expressed OC induce high glycolysis rate participate malignant invasion metastatic characteristics epithelial/mesenchymal transition (EMT). inhibitors suppress migration growth interfering effect. Proteoforms final structural functional forms gene/protein, canonical protein contains all proteoforms same gene. The complexity can be elucidated proteoformics. OC-specific proteoform might represent specific target therapeutic interventions against OC. In framework PPPM/3PM, warning prognosis biomarker. It important clarify molecular mechanisms metabolism. This review analyzes expression, function, OC, help

Abstract Leptomeningeal collaterals (LMCs) are crucial in mitigating the impact of acute ischemic stroke (AIS) by providing alternate blood flow routes when primary arteries obstructed. This article explores evolutionary pathophysiology LMCs, highlighting their critical function and genetic molecular mechanisms governing development remodelling. We address translational challenges applying animal model findings to human clinical scenarios, emphasizing need for further research validate emerging therapies—such as pharmacological agents, gene therapy mechanical interventions—in settings, aimed at enhancing collateral perfusion. Computational modelling emerges a promising method integrating experimental data, which requires precise parameterization empirical validation. introduce ‘Evolucollateral Dynamics’ hypothesis, proposing novel framework that incorporates biology principles into therapeutic strategies, offering new perspectives on circulation. hypothesis emphasizes role predispositions environmental influences circulation, may strategies optimize treatment outcomes. Future must incorporate data create robust protocols, thereby maximizing potential LMCs improving outcomes patients.
