Background: Non-alcoholic fatty liver disease (NAFLD) is increasing worldwide. A main cause the obesogenic, so-called Western lifestyle. NAFLD follows a long, unperceived course, and ends potentially fatally. Early diagnosis of aggressive subtypes saves lives. So far, non-invasive means detection are limited. better understanding pathogenic interplay among insulin resistance, immune inflammation, microbiome, genetic background important. Metabolomics may give insight into these interlaced processes. Methods: In this study, we measured bile acids (BA) in plasma adult alcohol-associated (ALD) patients healthy controls with targeted mass spectrometry. We focused on gender-related acid production pathology ALD. Results: Compared to controls, women had significantly higher concentrations total BA, primary cholic (CA), chenodeoxycholic (CDCA), glycine-conjugated, non-12-a-OH BA. Concerning subtypes, glycocholic (GCA) glycochenodeoxycholic (GCDCA), BA were elevated NAFLD. contrast, men no altered fractions. However, GCA, glycodeoxycholic (GDCA), glycolithocholic (GLCA), lithocholic (LCA), taurolithocholic (TLCA), tauroursodeoxycholic (TUDCA) elevated, while CA was decreased. NAFLD, except ursodeoxycholic (UDC), all correlated positively both sexes ELF score, ALD, only males showed significant correlations exceptive for UDC secondary free CA, CDCA, taurine conjugated, glycine 12-a-OH, cases high enhanced fibrosis (ELF) score above 9.8. additionally elevated. Between without NASH, found differences. Conclusion: Our data show gender-specific profiles markedly different patterns Women more severe cholestasis. Men compensate fat storage-driven dynamics, indicated by levels taurine-conjugated which associate beneficial metabolic functions.

Dietary fiber is a carbohydrate polymer with ten or more monomeric units that are resistant to digestion by human digestive enzymes, and it has gained widespread attention due its significant role in health improvement through regulating gut microbiota. In this review, we summarized the interaction between dietary fiber, microbiota, obesity, beneficial effects of on obesity modulation such as modifying selective microbial composition, producing starch-degrading improving barrier function, reducing inflammatory response, trimethylamine N-oxide, promoting production metabolites (e.g., short chain fatty acids, bile ferulic acid, succinate). addition, factors affecting microbiota composition (length chain, monosaccharide glycosidic bonds) were also concluded. Moreover, strategies for enhancing biological activity (fermentation technology, ultrasonic modification, nanotechnology, microfluidization) subsequently discussed. This review may provide clues deeply exploring structure–activity relationship antiobesity properties targeting specific

Nonalcoholic fatty liver disease (NAFLD) is the component of a cluster conditions, while its subtype, nonalcoholic steatohepatitis (NASH), emerges as potentially progressive disorder that harbors risk evolving into cirrhosis and culminating in hepatocellular carcinoma (HCC). NASH cardiovascular (CVD) have common factors, but compared to liver-related causes, most cause death patients CVD. Within pharmacological armamentarium, statins, celebrated for their lipid-modulating prowess, now garnered attention expansive therapeutic potential NASH. Evidence from plethora studies suggests statins not only manifest anti-inflammatory antifibrotic properties also impart multifaceted beneficial impact on hepatic health. In this review, we used “statin”, “NAFLD”, “NASH”, “CVD” major keywords conducted literature search using PubMed Web Science databases determine safety efficacy animals with NAFLD, mechanism statin therapy Simultaneously, reviewed important role intestinal microbiota NASH, it hoped will provide new insights modulating harmful inflammatory gut reducing systemic inflammation patients.

The metabolic implications in Alzheimer's disease (AD) remain poorly understood. Here, we conducted a metabolomics study on moderately aging Chinese Han cohort (n = 1397; mean age 66 years). Conjugated bile acids, branch-chain amino acids (BCAAs), and glutamate-related features exhibited strong correlations with cognitive impairment, clinical stage, brain amyloid-β deposition 421). These demonstrated synergistic performances across stages subpopulations enhanced the differentiation of AD beyond demographics Apolipoprotein E ε4 allele (APOE-ε4). We validated their eight data sets (total n 7685) obtained from Disease Neuroimaging Initiative (ADNI) Religious Orders Study Memory Aging Project (ROSMAP). Importantly, identified are linked to blood ammonia homeostasis. further confirmed elevated level through development 1060). Our findings highlight as emphasize metabolite-mediated disturbance its potential signature therapeutic target for AD.

Gut barrier is not only part of the digestive organ but also an important immunological for hosts. The disruption gut can lead to various diseases such as obesity and colitis. In recent years, traditional Chinese medicine (TCM) has gained much attention its rich clinical experiences enriched in thousands years. After orally taken, TCM interplay with microbiota. On one hand, modulate composition function other microbiota transform compounds. metabolites produced during actions these interplays exert noticeable pharmacological effects on host especially barrier. Recently, a large number studies have investigated repairing fortifying barriers from perspective metabolites. However, no review summarized mechanism behand this beneficiary TCM. review, we first briefly introduce unique structure specific Then, summarize interactions relationship amidst microbiota, Further, regulative mechanisms including physical barrier, chemical microbial At last, discuss that are associated destruction ulcerative colitis type 2 diabetes.
