Glycolysis-derived lactate was identified as substrate for histone lactylation, which has been regarded a significant role in transcriptional regulation many tissues. However, the of lactylation metabolic center, hypothalamus, is still unknown. Here, we show that hypothalamic pro-opiomelanocortin (POMC) neuron-specific deletion family with sequence similarity 172, member A (Fam172a) can increase and protect mice against diet-induced obesity (DIO) related disorders. Conversely, overexpression Fam172a POMC neurons led to an obesity-like phenotype. Using RNA-seq CUT&Tag chromatin profiling analyzes, find knockdown activates glycolytic process increases peptidylglycine α-amidating monooxygenase (PAM), affects synthesis α-MSH, via H4K12la (histone lactylation). In addition, pharmacological inhibition production clearly abrogates anti-obesity effect PFKO (POMC-Cre, Fam172aloxP/loxP, knockout). The prevalence of female-to-male (FtM) transgender individuals has risen recently, yet the effects cross-sex hormone therapy on volumetric differences in hippocampal subfields remain poorly understood. This study aimed to evaluate gray matter (GM) volume, especially focusing subfields, FtM and premenopausal cisgender women. Seventeen who had undergone hysterectomies were receiving testosterone before participating this study, along with 20 women, underwent a single session T1-weighted magnetic resonance imaging (MRI). group significantly higher free-testosterone (free-T) levels lower estradiol compared women (p < 0.001). In voxel-wise analysis, showed larger GM volumes caudate nucleus, hypothalamus, thalamus 0.01, FWE-corrected). More specifically, right subiculum volume was 0.05, Bonferroni-corrected), these positively correlated free-T (r = 0.34, p 0.04). revealed specific subfield testosterone-treated when group.
