Lung cancer is the most commonly diagnosed and one that causes deaths worldwide, so there a need for therapies improve survival rates. Products derived from marine organisms are source of novel potent antitumor compounds, but they present great obstacle their obtaining natural environment problems associated with synthesis biological effects chemical analogues. In this work, Bengamide analogue (Bengamide II) was chemically synthesized in vitro vivo studies were performed to determine its activity mechanisms action. It shown have antiproliferative lung lines 2D 3D models. addition, II-treated cells showed G2/M G0/G1 cell cycle arrest, together decrease proliferation marker Ki67. As mechanism action, treatment increased LC3-II expression production acidic vesicles signaling autophagy. II caspase-3 activation DNA fragmentation related apoptosis. Furthermore, reduction VEGFA expression, angiogenesis, also observed. markedly reduced tumor volume metastases increasing survival. Additionally, it revealed no systemic toxicity models at therapeutic doses used, which essential future clinical use. Taken together, bengamide II, promising drug showing relevant significant safety.
