Abstract The existence of the neural control mast cell functions has long been proposed. Mast cells (MCs) are localized in association with peripheral nervous system (PNS) and brain, where they closely aligned, anatomically functionally, neurons neuronal processes throughout body. They express receptors for regulated by various neurotransmitters, neuropeptides, other neuromodulators. Consequently, modulation provided these neurotransmitters neuromodulators allows MC involvement pathogenesis cell–related disease states. Recently, roles individual neuropeptides regulating actions have investigated extensively. This review offers a systematic recent advances our understanding contributions to activation pathological implications this regulation on states, though full extent which such influences health is still unclear, complete mechanisms underlying lacking. Future validation animal vitro models also needed, incorporates integration microenvironment-specific complex, multifaceted cross-talk between signals. Moreover, new biological agents directed against neurotransmitter that can be used therapeutic intervention need more specific, will reduce their ability support inflammatory responses enhance potential protecting pathogenesis.

Migraine stands as one of the most disabling neurological conditions worldwide. It is a disorder great challenge to study given its heterogeneous representation, cyclic nature, and complexity neural networks involved. Despite this, clinical preclinical research has greatly benefitted from use nitric oxide donor, nitroglycerin (NTG), model this disorder, dissect underlying mechanisms, facilitate development screening effective therapeutics. NTG capable triggering migraine attack, only in migraineurs or patients with history inducing migraine-like phenotypes rodent models. however unclear what extent NO, breakdown product, determinant factor pathophysiology migraine, importantly, whether it really does translation bench bedside, vice-versa. Protein S-nitrosation (SNO), a posttranslational modification (PTM) of cysteine (Cys) residues elicited by nitric oxide (NO), regulates wide range protein functions. As crucial form redox-based signaling NO, SNO contributes significantly to the modulation physiological functions, and imbalance is closely linked pathophysiological processes. Site-specific identification critical for understanding underlying molecular mechanisms function regulation. Although careful verification needed, data containing numerous functional proteins are potential research direction druggable target drug discovery. Undoubtedly, SNO-related meaningful not only development NO donor drugs but also classic target-based design. Herein, we provide comprehensive summary SNO, including its origin transport, identification, function, contribution Importantly, propose new views develop novel therapies based on SNO-sourced targets.

Migraine is a serious central nervous system disease with high incidence rate. Its pathogenesis very complex, which brings great difficulties for clinical treatment. Recently, many studies have revealed that mitochondrial dysfunction may play key role in migraine, affects the hyperosmotic of Ca 2+ , excessive production free radicals, decrease membrane potential, imbalance mPTP opening and closing, oxidative phosphorylation level, leads to neuronal energy exhaustion apoptosis, finally lessens pain threshold migraine attack. This article mainly introduces cortical spreading depression, then damages related function mitochondria, migraine. Clinically, calcitonin gene-related peptide antagonising drugs are recognized as effective in migraine treatment, but their site of action is debated. Only a small fraction these compounds pass the blood-brain barrier and accesses central nervous system. Regardless, it has been argued that system action. Here, we test this hypothesis by bypassing through intracerebroventricular injection drugs. We used glyceryl trinitrate (GTN) mouse model, which well validated its response to specific drugs. The receptor antagonist olcegepant monoclonal antibody ALD405 were administered either intraperitoneally or intracerebroventricularly. outcome measure was cutaneous mechanical allodynia.Mice given + GTN on day 1 had mean 50% withdrawal threshold 1.2 g contrast mice receiving placebo GTN, 0.3 (p < 0.001). Migraine headaches are highly prevalent, affecting 15% of the population. However, despite many studies to determine this disease's mechanism and efficient management, its pathophysiology has not been fully elucidated. There suggested hypotheses about possible mediating role mast cells, immunoglobulin E, histamine, cytokines in disease. A higher incidence disease allergic asthma patients, reported by several studies, indicates brain cells located around vessels The more specifically within dura can affect trigeminal nerve cervical or sphenopalatine ganglion, triggering secretion substances that cause migraine. Neuropeptides such as calcitonin gene-related peptide (CGRP), neurokinin-A, neurotensin (NT), pituitary adenylate-cyclase-activating (PACAP), substance P (SP) trigger response, they secrete pro-inflammatory vasodilatory molecules interleukin-6 (IL-6) vascular endothelial growth factor (VEGF) a selective result corticotropin-releasing hormone (CRH) secretion. This stress contributes migraine intensifies it. Chronic pain is a major burden for the society and remains more prevalent severe in females. The presence of chronic linked to persistent alterations peripheral central nervous system. One main types transducers are transient receptor potential channels (TRP), also known as thermoTRP channels, which intervene perception hot cold external stimuli. These especially TRPV1, TRPA1 TRPM8, have been subjected profound investigation because their role thermosensors implication acute pain. Surprisingly, sensitivity endogenous signaling has far less studied. Cumulative evidence suggests that function these may be differently modulated males females, part through sexual hormones, this could constitute significant contributor sex differences Here, we review exciting advances pharmacology females two paradigmatic with strong component: migraine chemotherapy-induced neuropathy (CIPN). possibilities druggability offered by differential exploitation men women represent development opportunity will lead increment armamentarium analgesic medicines personalized treatment.
