ABSTRACT: Omega-3 fatty acids (OFAs) are essential macromolecules which frequently used to provide nutritional support in cancer patients. They have been recognised as immunonutrients, they play important role strengthening the immune responses against various disease including cancer. OFAs such eicosapentaenoic acid (EPA) and docosahexaenoic (DHA) shown great promise prevention its management. Epidemiological studies demonstrated a correlation between fish consumption reduced risk, evidence suggests that increased is linked lower risk of cancer, potentially due high levels bioactive fish. Experimental revealed capable modulating cell signalling pathways, gene expression influencing membrane composition, can promote apoptosis inhibition proliferation. Moreover, antioxidant property has reported oxidative stress-induced DNA damage. Apart from anticancer properties also good results managing related complications inflammation, gastrointestinal reactions, anorexia-cachexia syndrome. Several showed their efficacy relieving associated cachexia anorexia syndrome with significant improvements weight loss. OFA supplementation antidepressant enhanced well-being individuals. Despite promising effects many challenges remain like optimal dose determination, variable impact across types risks intake. Further large scale randomized control trials (RCTs) needed assisted treatment.

Enhanced inflammatory and immune responses have been observed in patients with major depressive disorder, pointing to anti-inflammatory substances as potential seeds for developing novel antidepressants. Omega-3 polyunsaturated fatty acid metabolites, such resolvin D E series, maresins, protectins (collectively known specialized pro-resolving mediators) demonstrate effects. This study examined the antidepressant-like effects of maresin-1 (MaR1) on lipopolysaccharide (LPS)-induced depression-like behaviors mice. Using tail suspension test (TST) forced swim (FST), we assessed 26 28 h after intraperitoneal injection LPS (0.8 mg/kg), respectively. An open field (OFT) was also conducted evaluate locomotor activity 24 injection. Intracerebroventricular (i.c.v.) MaR1 (10 ng/mouse) immediately challenge mitigated increased immobility time TST FST, without affecting OFT, indicating preventive LPS-induced behaviors. Neohesperidin, a citrus flavonoid, shows potential for activating the mechanistic target of rapamycin complex 1 (mTORC1). Here, antidepressant-like effect neohesperidin was examined in male ICR mice (naïve and treated repeatedly with prednisolone, synthetic glucocorticoid, which induces depression-like behavior). Oral administration exerted an forced swim test h post-treatment, naïve mice; this no longer observed at 24 h. Neohesperidin also reversed prednisolone-induced behavior. This blocked by infusing rapamycin, mTORC1 inhibitor, into medial prefrontal cortex. may rapidly produce effect.

Abstract Specialized pro‐resolving lipid mediators play key functions in the resolution of acute inflammatory response. Herein, we elucidate stereochemical structure new 4S,5R‐RCTR1, a cysteinyl‐resolvin, recently uncovered human leukocytes incubated with 4S,5S‐epoxy‐resolvin intermediate, using liquid chromatography–tandem mass spectrometry (LC–MS/MS) and ultra‐violet (UV) spectrophotometry. With this approach, physical properties mediator prepared by total organic synthesis were matched to enzymatically produced biogenic material. In addition, confirmed potent biological actions 4S,5R‐RCTR1 M2‐like macrophage phagocytosis live bacteria, efferocytosis apoptotic neutrophils, erythrophagocytosis senescent red blood cells concentration‐dependent manner from 0.1 10 nM. Taken together, these results establish complete stereochemistry as 5R‐glutathionyl‐4S,17S‐dihydroxy‐6E,8E,10Z,13Z,15E,19Z‐docosahexaenoic acid give evidence its novel bioactivities phagocyte responses. Abstract Vascular endothelial growth factor (VEGF) signaling is known to be involved in the antidepressant‐like effects of conventional antidepressants, such as desipramine (DMI), a tricyclic antidepressant, and fluoxetine (FLX), selective serotonin reuptake inhibitor; however, precise role neuronal VEGF mediating these remains unclear. Using mice with excitatory neuron‐specific deletion its receptor, fetal liver kinase 1 (Flk‐1) forebrain, we examined forebrain or Flk‐1 on repeated DMI chronic FLX administration forced swim test (FST). Repeated intraperitoneal (i.p.) injections (10, 10, 20 mg/kg at 24, 4, h before FST, respectively) significantly decreased immobility control mice; this effect was completely blocked deletion. Although treatment (18 mg/kg/day, i.p.) did not impact day after 22nd injection, reduced preswim 23rd injection. However, deletion, increased failed produce effects. Collectively, findings indicate that VEGF–Flk‐1 contributes actions antidepressants.

Ketamine, an N-methyl-D-aspartate receptor antagonist, elicits swift antidepressant effects even in subjects with treatment-resistant depression. Nonetheless, owing to the serious adverse associated ketamine, including psychotomimetic effects, development of safer rapid-acting antidepressants is imperative. The elucidation mechanisms underlying ketamine will facilitate advancement these alternative treatments. Previous preclinical studies have indicated that properties are mediated by activity-dependent release brain-derived neurotrophic factor (BDNF) and subsequent activation mechanistic target rapamycin complex 1 (mTORC1) medial prefrontal cortex (mPFC). Our research has demonstrated exerts antidepressant-like inducing vascular endothelial growth (VEGF) insulin-like factor-1 (IGF-1) mPFC. Furthermore, our recent findings revealed resolvins (RvD1, RvD2, RvE1, RvE2, RvE3), which bioactive lipid mediators derived from docosahexaenoic eicosapentaenoic acids, exhibit rodent models. Notably, RvD1, RvE1 require mTORC1 activation. Moreover, intranasal administration rapid through BDNF VEGF mPFC hippocampal dentate gyrus (DG), as well mPFC, albeit not DG. These strongly suggest resolvins, particularly hold promise prospective candidates for novel, safer, antidepressants.

モノアミン仮説に基づく従来の抗うつ薬は即効性がなく,3割以上の患者が治療抵抗性を示す.一方,NMDA受容体阻害薬ケタミンは,治療抵抗性うつ病患者に即効性かつ持続性の抗うつ作用を示すことが明らかにされている.しかし,ケタミンには依存性や統合失調症様症状といった重大な副作用があるため,抗うつ薬としての使用には大きな制約を伴う.そのため,より安全性の高い新たな即効性抗うつ薬の開発が求められている.我々は,その候補としてドコサヘキサエン酸やエイコサペンタエン酸の活性代謝物であるレゾルビン類に注目し,これらがうつ病モデルマウスに対して抗うつ様作用を示すことを明らかにしてきた.本稿では,レゾルビン類のなかで最も低用量で抗うつ様作用を示したレゾルビンE1(RvE1)に焦点を当て,その抗うつ特性と作用メカニズムについて紹介する.まずRvE1側脳室内投与が,リポポリサッカライド誘発うつ病モデルマウスに対して抗うつ様作用を示し,この作用にケメリン受容体ChemR23活性化が関与する可能性を明らかにした.RvE1側脳室内投与は,慢性疼痛モデルマウスや三環系抗うつ薬の急性投与に抵抗性を示すプレドニゾロン反復投与誘発うつ病モデルマウスにも抗うつ様作用を示した.また,RvE1の抗うつ様作用には,内側前頭前野(mPFC)や背側海馬歯状回(DG)が関与することを見出した.さらに,RvE1経鼻投与が,ケタミンと類似のメカニズムで抗うつ様作用を示すことを明らかにした.すなわち,RvE1はmPFCおよび背側DGにおける活動依存的なBDNFおよびVEGF遊離亢進と,mPFCにおけるmechanistic target of rapamycin complex 1活性化を介して即効性かつ持続性の抗うつ様作用を示す.以上の知見から,RvE1が新しい即効性抗うつ薬のリード化合物になると期待される.
