Atopic dermatitis (AD) is a multifaceted inflammatory skin condition characterized by the involvement of various cell types, such as keratinocytes, macrophages, neutrophils, and mast cells. Research indicates that flavonoids possess anti-inflammatory properties may be beneficial in management AD. However, investigation glycoside forms for anti-AD therapy limited. We aimed to assess ability quercetin-3-O-glycosides treating AD-like lesions through silico-, cell-, animal-based platforms. The glycosylated flavonols quercitrin, isoquercitrin, rutin were used this study. also tried understand influence glycone type on bioactivity delivery glycosides. glycosides effectively reduced overexpression proinflammatory effectors interleukin (IL)-6, chemokine (C-X-C motif) ligand (CXCL)1, CXCL8, regulated upon activation normal T expressed secreted (RANTES), thymus activation-regulated (TARC) activated keratinocytes. This reduction could due inhibition extracellular signal-regulated kinase (ERK) p38 phosphorylation. Isoquercitrin (but not quercitrin rutin) arrest upregulated IL-6 CCL5 macrophage model. significantly prevented histamine release from RBL-2H3 absorption examination showed greater permeation isoquercitrin than with dual sugar moieties smaller molecular volume higher lipophilicity. deposition was enhanced about 11-fold stripped delipidized skins, which mimicked AD lesions. vivo dinitrochlorobenzene (DNCB)-induced mouse model demonstrated less erosion, scaling, epidermal hyperplasia after topical treatment. concentration cytokines/chemokines lesion decreased isoquercitrin. These effects similar those tacrolimus ointment. immunohistochemistry (IHC) displayed hyperproliferation immune infiltration results indicated quercetin provide an efficient safe way treat inflammation.

Allergic contact dermatitis (ACD) is a skin condition characterized by inflammation resulting from hypersensitivity upon with certain allergens. Although ACD an immune-mediated pathomechanism, the involvement of nervous system in this has increasingly been considered, particularly amplification and persistence inflammation. This paper aims to present comprehensive overview mechanisms involved neurogenic ACD, focusing on role sensory neurons, release neuropeptides, their interaction immune cells, potential therapeutic implications related pathways, diversified age gender. Innovative therapies for including topical formulations, may target mass-bound X2 G-protein-coupled receptor (MRGPRX2) endocannabinoid systems.

Chronic pruritus (≥ 6 weeks) is a frequent symptom in atopic diseases, with phenotypes ranging from non-lesional skin to inflammatory diseases like dermatitis. Data on patients' needs and treatment goals depending the phenotype disease burden are limited. This study aimed analyse impact of distinct chronic goals. Another objective was investigate whether influences Patient-reported outcomes 1,086 adult patients (n = 529 dermatitis, n 557 diathesis) were analysed age- gender-matched (mean age 49.7 ± 19.0 years; 605 female [55.7%]), comparing intensity (Numeric Rating Scale), quality life (Dermatology Life Quality Index, ItchyQol), anxiety depression (Hospital Anxiety Depression patient (Patient Needs Questionnaire Patient Benefit Index-Pruritus). Although significantly higher dermatitis (prolonged duration, increased impairment, intensity), treat-ment both matched 92.6%. The most important no longer experience itching, find clear diagnosis therapy, have confidence therapy.

ABSTRACT Itch and pain are both mediated by small sensory fibres. Atopic dermatitis (AD) patients usually report stress‐induced flares, but the impact of stress on fibres in lesional non‐lesional skin remains inconclusive. This observational study assessed effect acute profiles subjects with AD ( n = 18) healthy controls (HC, 21). Participants completed clinical psychological questionnaires, quantitative testing was performed HC. Assessments were done before after Montreal Imaging Stress Task, an protocol. responses evaluated anxiety ratings, heart rate (HR) salivary cortisol (CORT). Cortisol binding globulin (CBG) quantified as indirect measure for circulating CORT. participants reported higher anxiety, depression perception than HR similar between groups, showed a blunted CORT response post‐stress lower CBG levels, suggesting altered regulation. Acute reduced cold sensitivity HC had no lesions. These findings indicate that effects depend condition emphasise alterations experienced patients.

Abstract Atopic dermatitis (AD) is still a demanding challenge in clinical practice. Type 2 inflammation the most common inflammatory pathway children and adolescents with AD. Anti‐inflammatory drugs, mainly corticosteroids (CS) immunomodulant agents are primary therapeutic approach to dampening type inflammation. However, AD patients may require long‐term high CS doses or drug combinations possibly significant adverse effects achieve maintain disease control. In this regard, advent of biologics constituted breakthrough managing condition. Atopic dermatitis (AD) is a chronic inflammatory skin disease with complex pathophysiology characterized by intense pruritus, often associated psychological stress and atopic non-atopic comorbidities that significantly reduce quality of life. The aspects AD the interaction between mind body via skin–brain axis have led to an interest in mind–body therapies (MBT). aim this article is, therefore, reinforce importance psychodermatological care AD. We performed focused literature review on holistic practices or integrative MBT AD, including education, cognitive behavioral therapy, habit reversal, meditation, mindfulness, hypnotherapy, eye movement desensitization reprocessing, biofeedback, progressive muscle relaxation, autonomous sensory meridian response, music massage, touch therapy. A multidisciplinary approach MBT, addition conventional pharmacologic antipruritic therapies, break itch–scratch cycle may improve outcomes well-being. Although there paucity rigorously designed trials, evidence shows potential benefits pain, stress, anxiety, depressive symptoms, sleep quality. Relaxation various interventions, such as reversal therapy for replacing harmful scratching massaging emollient ‘plus’, urge scratch, while education adherence therapies.
