Mathematical analysis of chimeric antigen receptor T-cell therapy for leukaemia using optimal control approach

Abstract Leukaemia is a type of malignant blood cancer, and the most common cause cancer-related death leukaemia. In this study, we present comprehensive mathematical analysis T cell-based cancer treatment. We reconstruct model for chimeric antigen receptors (CAR) cell therapy targeting leukaemia adding new compartmental term cytokine in model. create to simulate receptor five nonlinear differential equations (MM) leukaemia, which includes susceptible cells $${S}_{1}(t)$$ S 1 ( t ) , infected $${I}_{1}(t)$$ I $${C}_{1}(t)$$ C immune $${W}_{1}(t)$$ W $${C}_{2}(t)$$ 2 describe dynamics transmission disease. employ optimal control (OC) theory, as CAR impacts both normal cells. The optimized T-cell dosage serves pivotal role introduced into system controller denoted $${u}_{1}$$ u . Conversely, manage release syndrome that leads singularity, introduce another $${u}_{2}$$ This controller, tocilizumab, acts an immunosuppressive agent within system. have discussed simulation results two ways: one approach (using SDRE method), conventional (without control).
